摘  要:为了研究朝藿定A对破骨细胞及骨质疏松雄性小鼠的作用,本试验将0.01~1.00μmol/L朝藿定A用于正常小鼠骨髓单核细胞诱导形成破骨细胞,分别于7,14 d进行抗酒石酸酸性磷酸酶(tartrate resistant acid phosphatase,TRACP)染色和骨吸收陷窝染色;将5,10,20 mg/kg朝藿定A用于去势诱导的骨质疏松雄性小鼠,8周给药结束后处死小鼠并检测血清骨保护素(osteoprotegerin,OPG)和小鼠Ⅰ型胶原C端肽(C-terminal telopeptide of typeⅠcollagen,CTXⅠ)含量,采用Micro-CT对胫骨内部结构进行扫描,用HE染色观察股骨上端骺板软骨细胞和骨小梁。结果显示,在朝藿定A给药后,破骨细胞的形成和TRACP分泌减少,破骨细胞对骨薄片的吸收能力降低;朝藿定A给药组小鼠血清中OPG、CTXⅠ水平降低,小鼠骨小梁数目增加,骨微形态结构有所改善。结果表明,朝藿定A可以通过抑制破骨细胞的形成、分化和骨吸收作用,改善骨质疏松模型小鼠的骨微结构和血清骨转换标志物含量,起到骨保护作用。In order to study the effects of epimedin A on osteoclasts and osteoporotic male mice,0.01-1.00μmol/L epimedin A was used to induce osteoclasts isolated from bone marrow mononuclear cells of normal mice,and TRACP staining and bone resorption lacuna staining were performed on 7,14 d,respectively.Osteoporotic male mice were treated with 5,10,20 mg/kg epimedin A,and the mice were killed after 8 weeks of administration.The contents of osteoprotegerin(OPG)and C-terminal telopeptide of typeⅠcollagen(CTXⅠ)in serum were detected;the internal structure of tibia was scanned by micro-CT;the chondrocytes and trabeculae of femur were observed by HE staining.The results showed that after the administration of epimedin A,the formation of osteoclasts and the secretion of TRACP decreased,and the absorptive capacity of osteoclasts to bone slices reduced,the levels of OPG and CTXⅠin serum declined,the number of bone trabeculae increased,and the bone microstructure improved.In conclusion,epimedin A can protect bone formation by inhibiting serum bone turnover markers and osteoclast differentiation and bone resorption.
