摘 要:旨在探索感染猪繁殖与呼吸综合征病毒(porcine reproductive and respiratory syndrome virus,PRRSV)后引起继发性严重炎症反应的细菌性病原,以期为疾病的诊断、治疗和预防提供参考依据。从感染PRRSV的猪体内无菌采取胸腔积液,经菌落培养、革兰染色观察、生理生化鉴定、药物敏感性试验、16S rRNA序列比对分析和动物致病性试验,了解病原菌生长特性、菌落形态、生理生化特性、药物敏感性和致病性。结果显示,该分离菌株在普通琼脂平板呈圆形、白色小菌落,血平板呈α溶血,革兰染色为阳性球菌、呈两联或四联,能发酵葡萄糖、麦芽糖、乳糖、蔗糖,赖氨酸脱羧酶反应呈阳性,对头孢他啶、头孢呋辛、头孢西啶、苯唑西林高度敏感,对万古霉素、青霉素G不敏感。16S rRNA基因序列分析显示,该分离菌株与NCBI登录的绿色气球菌的同源性高达99%,小鼠攻菌试验表明该分离菌株具有致病性。本试验为进一步探索绿色气球菌的致病机制奠定前期基础,并为动物疾病的诊断、预防和治疗提供技术支撑和实践指导。The aim of this study is to identify the bacterial pathogens causing pleural effusion of pigs infected with porcine reproductive and respiratory syndrome virus(PRRSV),and analyze the physiological and biochemical characteristics,pathogenicity and genome-wide characteristics of the isolated pathogens.In order to understand the growth characteristics,colony morphology,physiological and biochemical characteristic,drug sensitivity and pathogenicity of the pathogen,bacterial colony culture,Gram staining observation,physiological and biochemical identification,drug sensitivity test,16 S rRNA sequence comparison and animal pathogenicity test were conducted.The results showed that the isolated strain was Gram positive cocci,it could form round,white,double or quadruple colony on ordinary agar plate,it could ferment glucose,maltose,lactose and sucrose.The lysine decarboxylase reaction of the isolated bacteria was positive and it was highly sensitive to ceftazidime,cefuroxime,cephalosporin and oxacillin and not sensitive to vancomycin and penicillin G.Sequence analysis of 16 S rRNA gene showed that the isolate shared 99%homology with Aerococcus viridans registered in NCBI database.The mouse challenge test showed that the isolated strain was pathogenic.This study laid a preliminary foundation for further exploring the pathogenic mechanism of Aeromonas aeruginosa,which can provide technical support and practical guidance for the diagnosis,prevention and treatment of animal diseases.
