摘 要:为了比较猪繁殖与呼吸综合症病毒(porcine reproductive and respiratory syndrome virus,PRRSV)HeN-3弱毒株经滴鼻和注射两种途径感染仔猪后诱导天然免疫因子产生的差异,经滴鼻和注射两种途径感染仔猪后,前腔静脉采集凝血与抗凝血,通过ELISA Kit检测血清中IFN-α蛋白水平;通过荧光定量PCR方法检测血清中病毒拷贝数及外周血淋巴细胞的细胞因子IFN-α、TNF-α、IL-1β的相对表达量并分析其差异;43 d后剖检仔猪,收集仔猪的肝脏、脾脏、肾脏制作病理切片,观察组织损伤情况。结果显示,两种途径感染仔猪的健康状况与对照组基本一致;注射组在3 d,滴鼻组在7 d出现病毒血症,注射组比滴鼻组的病毒血症持续时间长且血清中病毒拷贝数更高;两种途径感染仔猪外周血淋巴细胞中IFN-α的相对表达量先抑制后显著上调,血液中IFN-α蛋白水平在3,11,15 d显著高于对照组;两种途径感染仔猪的天然免疫因子IL-1β、TNF-α相对表达显著上调,但持续时间较短;两种途径感染仔猪的肝脏、脾脏、肾脏等组织器官未有明显病理变化。结果表明,PRRSV HeN-3弱毒株以滴鼻和注射两种方式感染仔猪,都未出现临床症状和病理变化,均能诱导产生一定水平的IFN-α,引起的天然免疫因子变化趋势相似。The purpose of this study is to compare the difference of innate immune factors induced by porcine reproductive and respiratory syndrome virus(PRRSV)HeN-3attenuated strain in piglets infected via intranasal drip or injection.After piglets were infected via nasal drip or injection,coagulated and anticoagulated blood were collected through anterior vena cava,and the level of interferon in serum was measured by ELISA and the relative expression levels of cytokines IFN-α,TNF-αand IL-1βin peripheral blood lymphocytes were measured by fluorescence quantitative PCR.The piglets were dissected 43dpost virus inoculation,the liver,spleen and kidney were collected to make pathological sections,and the tissue injury was observed.The results showed that,the health status of piglets infected via nasal drip or injection was basically the same as that of the control group;viremia occurred on the 3din the injection group and 7din the nasal drip group,the duration of viremia in the injection group was longer;the number of virus copies in serum was higher than that in the nasal drip group;the relative expression of IFN-αin peripheral blood lymphocytes of piglets infected by the two ways was initially inhibited and then significantly upregulated,and the IFN-αlevel in blood was significantly higher than that in the control group on 3,11,15d;the relative expression of innate immune factors IL-1βand TNF-αin piglets infected by nasal drip or injection was significantly up-regulated,but the duration was short;there were no obvious pathological changes in liver,spleen,kidney and other tissues and organs of piglets infected via nasal drip or injection.The results of this study indicated that piglets infected with PRRSV HeN-3attenuated strain via intranasal drip or injection did not show clinical symptoms and pathological changes,both infection routes could induce a certain level of IFN-αand the change trend of innate immune factors was similar.
