摘 要:为了探究中药多糖对代森锰锌(MCZ)诱导雌性小鼠氧化及卵巢毒性的缓解作用,将70只小鼠随机分为7组,每组10只。空白组每天分2次灌服0.2 mL去离子水,MCZ模型组每天灌服100 mg/kg MCZ和0.2 mL去离子水,中药多糖处理组每天灌服100 mg/kg MCZ及分别灌服200 mg/kg枸杞多糖(LBP)、黄芪多糖(APS)、甘草多糖(GP)、菟丝子多糖(CCP)和山药多糖(CYPS)。用药30 d后处死,收集血液、子宫和卵巢。统计子宫和卵巢指数;HE染色观察卵巢组织形态学变化;可见光法、TBA法和WST-1法分别检测血清中过氧化氢酶(CAT)、丙二醛(MDA)和超氧化物歧化酶(SOD)含量;放射性免疫法检测血清中卵泡刺激素(FSH)、黄体生成素(LH)和雌二醇(E2)水平;酶联免疫吸附法(ELISA)检测子代雌鼠卵巢内雌激素受体α(ERα)、雌激素受体β(ERβ)含量;实时荧光定量PCR检测卵巢内NQO1、GcLc、Gstt2、HO1和GPx2 mRNA转录水平。结果显示,MCZ可以降低CAT、SOD活力和MDA、FSH、LH含量及ERα、ERβ表达,升高E2含量,下调NQO1、GcLc、Gstt2、HO1 mRNA转录水平,上调GPx2 mRNA转录水平;LBP、APS、CCP、CYPS和GP可在一定程度上缓解MCZ的影响。本研究表明,MCZ可通过Keap1-Nrf2通路对小鼠造成损伤,而中药多糖干预Keap1-Nrf2通路缓解MCZ毒性,其中GP效果最佳。In order to explore the effect of traditional Chinese medicine polysaccharides on oxidation and ovarian toxicity induced by mancozeb(MCZ)in female mice,70 mice were randomly divided into 7 groups with 10 mice in each group(n=10).The blank group was fed with 0.2 mL deionized water twice a day,the MCZ model group was fed with 100 mg/kg MCZ and 0.2 mL deionized water,and the traditional Chinese medicine polysaccharide group was orally administrated with 100 mg/kg MCZ and 200 mg/kg Lycium barbarum polysaccharides(LBP),Astragalus polysaccharide(APS),Glycyrrhiza polysaccharide(GP),Cuscuta chinedsis polysaccharides(CCP)and Chinese Yam polysaccharide(CYPS).The mouse was killed after 30 d of treatment,and the blood,uterus and ovaries were collected.The indexes of uterus and ovary were counted,the histomorphological changes of ovary were observed by HE staining,the contents of catalase(CAT),malondialdehyde(MDA)and superoxide dismutase(SOD)in serum were detected by visible light method,TBA method and WST-1 method,respectively,and the levels of follicle stimulating hormone(FSH),luteinizing hormone(LH)and estradiol(E2)in serum were detected by radioimmunoassay.The contents of estrogen receptorα(ERα)and estrogen receptorβ(ERβ)in the ovaries of the offspring were detected by ELISA,and the mRNA transcription levels of NQO1,GcLc,Gstt2,HO1and GPx2in the ovaries were detected by qRT-PCR.The results showed that MCZ could decrease the activity of CAT,SOD and the content of MDA,FSH,LH and the expression of ERα,ERβ,increase the content of E2,down-regulate the mRNA transcriptional levels of NQO1,GcLc,Gstt2and HO1,and up-regulate the mRNA transcriptional level of GPx2,while LBP,APS,CCP,CYPS and GP could alleviate the effect of MCZ to some extent.This study shows that MCZ can cause damage to mice through Keap1-Nrf2pathway,while traditional Chinese medicine polysaccharides interfere with Keap1-Nrf2pathway to alleviate MCZ toxicity,among which GP has the best effect.
