摘  要:目的研究屋尘螨Derp1蛋白的性质,通过在线软件明确CD4~+T细胞和CD8~+T细胞抗原表位分布情况,并找出其优势表位,为尘螨过敏特异诊断和免疫治疗提供新思路和理论依据。方法通过ExPASy的ProtParam tool在线软件输入Derp1蛋白的氨基酸序列分析其理化性质;通过TMpred软件输入Derp1蛋白的氨基酸序列检测该蛋白质的跨膜区特征。经NCBI检索出Derp1基因及其编码氨基酸序列,登录NetMHCIIpan 3.2 Server在线软件输入Derp1基因序列,找出CD4~+T细胞抗原表位;分别登录NetMHC 4.0 server、SYFPEITHI、NetCTL 3种在线软件,用上述相同方法找出CD8+T细胞抗原表位。结合4种在线软件的预测结果,找出Derp1蛋白的优势表位。结果 Derp1基因开放阅读框长度为1 400 bp,编码320个氨基酸,Derp1共4 980个原子,相对分子质量为36 078.4,分子式为C_(1598)H_(2438)N_(444)O_(487)S_(13),等电点为5.56。Derp1蛋白存在螺旋跨膜区分别为1-23、125-148位氨基酸处,两处螺旋区的总评分为2 786。预测Derp1蛋白含有6条CD8~+T抗原表位,优势表位的序列分别为:262-270 GYQPNYHAV、115-123RQMRTVTPI、203-211 RPNAQRFGI、300-308 GYFAANIDL、249-257 RHYDGRTII和264-272 QPNYHAVNI。Derp1含有的CD4~+T细胞强结合表位为21条。总结在线软件预测的CD4~+T和CD8~+T细胞抗原表位,最终挑选出的优势表位肽为RQMRTVTPI。结论候选优势表位RQMRTVTPI将作为尘螨过敏特异诊断和免疫治疗的研究基础,为今后临床提高过敏性疾病的诊断及治疗提供理论依据。Objective To investigate the nature of house dust mite Derp1 protein,clarify the distribution of CD4+and CD8+T cells antigenic epitopes through online software and identify the dominant epitopes so as to provide new ideas and theoretical basis for the specific diagnosis and immunotherapy of dust mite allergy.Methods The amino acid sequence of Derp1 protein was entered into ProtParam tool online software of ExPASy to analyze the physicochemical properties,and into TMPred software to detect the characteristics of transmembrane region.Derp1 gene and the amino acid sequence encoded by the gene were subjected to the NCBI retrieval,while the former was entered into NetMHCIIpan 3.2 Server online software to find out the antigenic epitopes of CD4+T cells.The antigenic epitopes of CD8+T cells were identified by the above-mentioned method using three online software,NetMHC 4.0 server,SYFPEITHI and NetCTL.The dominant epitopes of Derp1 protein were identified by combination of prediction results of the four online software.Results The open reading frame of Derp1 gene was 1400 bp in length,which encoded 320 amino acids.Derp1 consisted of 4980 atoms,of which the relative molecular mass was 36078.4,the molecular formula was C1598H2438N444O487S13,and the isoelectric point was 5.56.The Derp1 protein had helical transmembrane regions at amino acids 1-23 and 125-148 with a total score of 2786,which was predicted to contain six CD8+T-antigen epitopes,and the sequences of dominant epitopes were 262-270 GYQPNYHAV,115-123 RQMRTVTPI,203-211 RPNAQRFGI,300-308 GYFAANIDL,249-257 RHYDGRTII and 264-272 QPNYHAVNI.Derp1 contained 21 strong binding epitopes for CD4+T cells.Based on the summarization of CD4+T and CD8+T cell antigenic epitopes predicted by the online software,the final dominant epitope peptide selected was RQMRTVTPI.Conclusion The candidate dominant epitope RQMRTVTPI will be served as the research basis for specific diagnosis and immunotherapy of dust mite allergy,which provides a theoretical basis for the improvement of clinica
