摘　　要：目的探讨上皮细胞钠离子通道(epithelial Na+channel,ENaC)各个亚基表达在维吾尔族子痫前期胎盘组织中的表达及意义。方法定期产检并分娩或终止妊娠的维吾尔族子痫前期孕妇122例,根据发病时间分为早发型(孕周<34周)子痫前期50例(早发型组)和晚发型(孕周≥34周)子痫前期72例(晚发型组);定期产检并足月分娩的正常孕妇125例为对照组,取各组孕妇的胎盘组织,采用实时荧光定量PCR法检测胎盘组织ENaCα、β、γ3个亚基mRNA相对表达量,采用Western blot法检测胎盘组织ENaCα、β、γ3个亚基蛋白相对表达量。结果3组胎盘组织SCNN1A、SCNN1G mRNA和其编码的ENaCα、γ蛋白相对表达量比较差异均无统计学意义(P>0.05)。早发型组孕妇胎盘组织SCNN1B mRNA(0.880±1.090)和其编码的ENaCβ蛋白(0.728±0.345)相对表达量均低于对照组(3.490±1.020、1.201±0.621)和晚发型组(3.99±2.12、1.424±0.134)(P<0.05),晚发型组与对照组比较差异均无统计学意义(P>0.05)。结论ENaCβ亚基表达在维吾尔族早发型子痫前期呈低表达,ENaCβ亚基可能成为预测早发型子痫前期的指标。Objective To investigate the expression and significance of each subunit of epithelial Na+channel(ENaC)in the placenta of Uygur patients with preeclampsia(PE).Methods In 122 Uygur PE pregnant women who delivered or terminated pregnancy after regular physical examination,50 women developed PE at gestational age<34 weeks(early-onset group)and 72 women developed PE at gestational age≥34 weeks(late-onset group).Another normal 125 pregnant women received regular physical examination and delivered termly(control group).The relative expression of ENaC subunit mRNA was detected by real-time fluorescence quantitative PCR,and the relative expression of ENaC subunit protein was detected by Western blot in the placental tissue in all groups.Results There were no significant differences in the relative expressions of SCNN1 A and SCNN1 G mRNAs and proteins as well as the relative expressions of ENaCαandγproteins among three groups(P>0.05).The relative expressions of SCNN1 B mRNA and ENaCβprotein were lower in early-onset group(0.880±1.090,0.728±0.345)than those in control group(3.490±1.020,1.201±0.621)and late-onset group(3.99±2.12,1.424±0.134)(P<0.05),and showed no significant differences between late-onset group and control group(P>0.05).Conclusion The subunit of ENaCβis lowly expressed in early-onset PE Uygur patients,and it may be a predictor of early-onset PE.
