摘 要:目的研究轮叶党参总皂苷对糖尿病大鼠心肌组织丙二醛(MDA)、肿瘤坏死因子α(TNF-α)、可溶性细胞间粘附分子-1(s ICAM-1)及可溶性血管细胞间粘附分子-1(s VCAM-1)表达的影响,并探讨其心肌保护作用。方法构建大鼠2型糖尿病心肌病变模型,实验分组:正常对照组(生理盐水)、模型对照组(生理盐水)、轮叶党参组(轮叶党参总皂苷80mg·kg^(-1)·d^(-1)),各组灌胃8周,末次给药后腹主动脉采血并分离心脏,酶联免疫吸附法(ELISA)检测超氧化物歧化酶(SOD)、MDA和TNF-α的含量,采用聚丙烯酰胺凝胶蛋白电泳检测sICAM-1及s VCAM-1的表达。结果与正常对照组比较,模型对照组及轮叶党参组SOD活性均明显降低(P<0.05),MDA、TNF-α、sICAM-1及sVCAM-1含量均明显升高(P<0.05)。轮叶党参组与模型对照组相比,SOD活性明显升高(P<0.05),MDA、TNF-α、sICAM-1及sVCAM-1含量明显下降(P<0.05)。结论轮叶党参总皂苷可以显著降低MDA、TNF-α、sICAM-1及sVCAM-1含量,有显著的心肌保护作用。Objective To study the effect of total saponins of Codonopsis lanceolata on the expression of MDA,TNF-α,s ICAM-1 and s VCAM-1 in myocardium of diabetic rats,and explore its myocardial protective effect.Methods The type 2 diabetic cardiomyopathy rats model were constructed and randomly divided into normal control group(normal saline),model control group(normal saline)and Codonopsis lanceolata group(total saponins of Codonopsis lanceolata 80 mg·kg^(-1)·d^(-1)),the three groups were given intragastric administration for 8 weeks.After the last administration,blood was collected from abdominal aorta and heart was separated.Superoxide dismutase(SOD),malondialdehyde(MDA)and tumor necrosis factorα(TNF-α)were assessed by enzyme-linked immunosorbent assay(ELISA).The expression of soluble intercellular adhesive molecule-1(s ICAM-1)and soluble vascular cell adhesive molecule-1(s VCAM-1)were detected by polyacrylamide gel protein electrophoresis.Results Compared with normal control group,SOD activity in model control group and Codonopsis lanceolata group decreased significantly(P<0.05),while MDA,TNF-α,s ICAM-1 and s VCAM-1 contents increased significantly(P<0.05).Compared with model control group,SOD activity in Codonopsis lanceolata group increased significantly(P<0.05),while MDA,TNF-α,sICAM-1 and s VCAM-1 contents decreased significantly(P<0.05).Conclusion Total saponins of Codonopsis lanceolata can significantly reduce the contents of MDA,TNF-α,s ICAM-1 and s VCAM-1,and has significant myocardial protection effect.
