摘 要:为明确产紫篮状菌(Talaromycespurpureogenus)Q2菌株抑制苦瓜枯萎病病原菌尖镰孢菌苦瓜专化型(Fusariumoxysporumf.sp.momordicae)SG-15菌株生长机制,利用共培养法测定菌株Q2对菌株SG-15菌丝形态和分生孢子活性的影响,同时采用转录组学技术分析菌株SG-15诱导下产紫篮状菌Q2菌株的基因表达差异。结果显示,在菌株Q2的胁迫下,SG-15生长受到显著抑制,菌丝体细胞形态遭破坏,细胞内结构紊乱,细胞膜通透性受到严重破坏。在病原菌SG-15诱导下,菌株Q2的β-1,3-葡聚糖酶和几丁质酶产生能力及其相关基因的相对表达水平升高,同时,菌株Q2基因组中部分次级代谢产物相关基因的相对表达水平也升高。本研究结果表明,菌株Q2对尖镰孢菌生长的抑制作用是一个综合的作用机制,菌株Q2产生的β-1,3-葡聚糖酶和几丁质酶参与了菌株Q2对尖镰孢菌生长的抑制作用。In order to determine the inhibitory mechanism of Talaromyces purpureogenus strain Q2 to the growth of Fusarium oxysporum f.sp.momordicae strain SG-15,the co-culture method was adopted to detect the effects of Q2 strain on the mycelial morphological and conidia vitality of SG-15.At the same time,DEGs were identified to analyze the differential expression of strain Q2 between pure culture and co-culture with strain SG-15 according to RNA sequencing.The results showed that the strain Q2 could significantly inhibit the growth of SG-15;the cell morphology of mycelium were destroyed,the structure of cells became more irregular,and the cell membranes were seriously damaged.The production ofβ-1,3-glucanase and chitinase and the relative expression levels of their related genes in strain Q2 increased after induced by strain SG-15.Meanwhile,the results of transcriptome also confirmed that the relative expression level of genes associated with some secondary metabolite of strain Q2 increased.In conclusion,the inhibitory mechanism of Talaromyces purpureogenus strain Q2 against F.oxysporum was a comprehensive mechanism,theβ-1,3-glucanase and chitinase produced by strain Q2 involved in the inhibition effects on growth of F.oxysporum.
