摘 要:目前有关文献报道中是使用Chiralpak AD-H色谱柱测试氟苯尼考光学异构体[1],此色谱柱检测出的氟苯尼考光学异构体,其实是甲砜霉素峰,该色谱柱填料无法分离氟苯尼考光学异构体与甲砜霉素。该方法没有考虑到氟苯尼考样品中残留的甲砜霉素杂质,甲砜霉素与氟苯尼考光学异构体在此条件下是无法完全分离,导致氟苯尼考光学异构体无法准确检出。方法:采用HPLC法,色谱柱为大赛璐手性柱Chiralpak IC,正已烷—乙醇的混合液为流动相,检测波长为225 nm,柱温25℃,流速1.0 mL/min。结果:氟苯尼考、氟苯尼考光学异构体、甲砜霉素间的分离度良好,纯度系数均大于0.99,定量检测限度对应为0.03%、0.03%。结论:此方法便捷、专属性强、重现性好,可用于氟苯尼考光学异构体含量的检测。At present,relevant literature reports are using Chiralpak AD-H chromatographic column to test the optical isomers of florfenicol.The optical isomers of florfenicol detected by this column are actually thiamphenicol peaks.The column packing cannot separate the optical isomers of Florfenicol and Thiamphenicol.This method does not take into account the residual thiamphenicol impurities in the florfenicol sample.The optical isomers of thiamphenicol and florfenicol cannot be completely separated under this condition,resulting in the optical isomers of florfenicol.Cannot be detected accurately.Method:HPLC method was used,the chromatographic column was Daicel chiral column Chiralpak IC,the mixture of n-hexane and ethanol was the mobile phase,the detection wavelength was 225 nm,the column temperature was 25℃,and the flow rate was 1.0 mL/min.Results:The resolution of florfenicol,optical isomers of florfenicol and thiamphenicol was good,the purity coefficients were all greater than 0.99,and the quantitative detection limits were 0.03%and 0.03%,respectively.Conclusion:This method is convenient,specific,and reproducible,and can be used to detect the content of florfenicol optical isomers.
