摘 要:目的对青楷槭的二氯甲烷和乙酸乙酯萃取部位的化学成分进行研究,并考察青楷槭的乙醇总提取物、各萃取部位及分离得到的单体化合物的抗氧化活性。方法采用乙醇加热回流法得到青楷槭茎的乙醇总提取物,蒸馏水制成悬浮液,依次用二氯甲烷、乙酸乙酯进行萃取。利用正、反相硅胶及葡聚糖凝胶柱层析等分离技术进行分离,通过1H-NMR及13C-NMR等波谱方法对化合物的结构进行鉴定。利用DPPH法对青楷槭的粗提取物及分离得到的单体化合物进行体外抗氧化活性测试。结果醇粗提取物、二氯甲烷层、乙酸乙酯层和水层的IC50值分别为11.64、39.45、8.46和53.32 mg/L。从青楷槭的二氯甲烷层(1~7)和乙酸乙酯层中(8~15)分离得到15个单体化合物。化合物8~15的IC50值分别为23.40、15.87、14.84、32.52、34.84、31.25、14.14和9.92μmol/L。结论青楷槭茎的15个化合物中1、2、4、5为首次得到。青楷槭茎醇粗提物、乙酸乙酯层及其中分离的化合物具有良好的抗氧化活性。Objective To isolate and identify the chemical constituents from dichloromethane and ethyl acetate layer of the stems of Acer tegmentosum Maxim..To study the antioxidant activities of the total ethanol extract,the extract layers and the compounds of Acer tegmentosum.Methods The total ethanol extract from the stem of Acer tegmentosum was obtained by ethanol heating reflux method.The suspension was made from distilled water and extracted with dichloromethane and ethyl acetate in turn.The compounds were isolated by positive,reverse phase silica gel and Sephadex column chromatography.The structures of the compounds were identified by 1H-NMR and 13C-NMR spectroscopy.DPPH method was used to test the antioxidant activity of crude extract and isolated monomer compounds of Acer tegmentosum in vitro.Results The IC50 values of crude alcohol extract,dichloromethane layer,ethyl acetate layer and water layer were 11.64,39.45,8.46 and 53.32 mg/L,respectively.Fifteen compounds were isolated from dichloromethane layer(1-7)and ethyl acetate layer(8-15)of Acer tegmentosum.The IC50 values of compounds 8-15 were 23.40,15.87,14.84,32.52,34.84,31.25,14.14 and 9.92μmol/L,respectively.Conclusion Compounds 1,2,4 and 5 are obtained for the first time from the stems of Acer tegmentosum.The ethanol extract,ethyl acetate layer and the compounds isolated from the stem of Acer tegmentosum showed good antioxidant activity.
