摘  要:目的本研究观察单侧前庭功能丧失大鼠及经不同剂量天麻素治疗后前庭内侧核中GLU-IR、GFAP、GABA B2表达的变化。方法将99只SD大鼠随机分为空白对照组(KBDZ组,n=9)、假手术对照组(JSSDZ组,n=18)、模型对照组(MXDZ组,n=18)、模型治疗组(MXZL组,n=54),KBDZ组正常饲养,JSSDZ组鼓室内注射生理盐水,MXDZ组及MXZL组鼓室内注射氯仿(50%)。造模成功后,再次将MXZL组大鼠随机分为天麻素高、中、低三组各18只,(TMSGJL组,TMSZJL组、TMSDJL组),分别连续腹腔注射3日高、中、低剂量天麻素(50 mg·kg^(-1)·d^(-1)、75mg·kg^(-1)·d^(-1)、100 mg·kg^(-1)·d^(-1))。观察前庭内侧核中GLU-IR、GFAP、GABA B2的表达。结果MXDZ组、MXZL组与JSSDZ组、KBDZ组比较GLU-IR表达下调,GFAP、GABA B2的表达上调。天麻素治疗后,MXZL组与MXDZ组比较:GLU-IR、GABA B2的表达上调;GFAP的表达明显下调。结论单侧前庭功能丧失可导致前庭内侧核中GLU-IR表达下调,GFAP、GABA B2的表达明显上调。经天麻素治疗后,GLU-IR、GABA B2的表达明显上调,GFAP的表达明显下调,均为天麻素高剂量组最为明显。说明天麻素能够加速中枢代偿,为今后防治前庭神经损伤提供新思路。Objective The aim of this study was to investigate the expression of Glu-IR,GFAP and gabab2 in the medial vestibular nucleus of rats with unilateral vestibular dysfunction and treated with different doses of gastrodin.Methods A total of 99 SD rats were randomly divided into blank control group(KBDZ group,n=9),sham operation control group(jssdz group,n=18),model control group(mxdz group,n=18)and model treatment group(mxzl group,n=54).KBDZ group was fed normally,jssdz group was injected with normal saline,mxdz group and mxzl group were injected with chloroform(50%).After successful modeling,rats in mxzl group were randomly divided into three groups(tmsgjl group,tmszjl group and tmsdjl group)with 18 rats in each group.High,medium and low doses of gastrodin(50 mg·kg^(-1)·D^(-1),75 mg·kg^(-1)·D^(-1)and 100 mg·kg^(-1)·D^(-1))were injected intraperitoneally for 3 days.The expression of Glu-IR,GFAP and gabab2 in medial vestibular nucleus was observed.Results Compared with jssdz group and KBDZ group,the expression of Glu-IR in mxdz group and mxzl group was down regulated,while the expression of GFAP and gabab2 was up-regulated.After gastrodin treatment,compared with mxdz group,the expression of Glu-IR and gabab2 were up-regulated in mxzl group;The expression of GFAP was significantly down regulated.Conclusion Unilateral vestibular dysfunction resulted in down-regulation of Glu-IR expression and up-regulation of GFAP and gabab2 expression in medial vestibular nucleus.After treatment with gastrodin,the expression of Glu-IR and gabab2 were significantly up-regulated,while the expression of GFAP was significantly down regulated,which was the most obvious in the high-dose gastrodin group.It is suggested that gastrodin can accelerate the central compensation and provide a new idea for the prevention and treatment of vestibular nerve injury in the future.
