摘 要:目的探讨不同剂量的知母黄柏药对(ZB)对高脂饲料诱导肥胖症大鼠的降脂作用及其可能的作用机制。方法雄性SD大鼠62只,随机分为正常组12只,造模组52只。正常组给予普通饲料喂养,造模组以高脂饲料喂养24周;造模第21周,将造模组大鼠随机分为模型组,知母黄柏药对高、中、低剂量组(3.00,1.00,0.33g·kg^(-1)·d^(-1)),盐酸吡格列酮阳性药组(5.25mg·kg^(-1)·d^(-1)),每组各10只。正常组与模型组灌服等体积蒸馏水,其他实验组相应处理,持续22天,于24周末处理大鼠,取血清及肝组织,测量血清甘油三酯(TG),胆固醇(CHO)及总胆汁酸(TBA)水平变化,观察肝组织病理变化(HE染色)。结果与正常组比较,模型组大鼠血清TG、CHO含量均显著升高(P<0.01),血清TBA含量也显著升高(P<0.05);与模型组比较,知母黄柏高剂量组及知母黄柏中剂量组TG、TBA水平均显著降低(P<0.01),盐酸吡格列酮组TG水平显著降低(P<0.05),知母黄柏低剂量组和盐酸吡格列酮组TBA水平显著降低(P<0.01),组织病理显示,知母黄柏药对使各剂量组大鼠肝细胞脂肪变性、气球样变及炎症的程度明显减轻。结论知母黄柏药对不同剂量对肥胖症均具有良好的预防和治疗的作用,其机制可能与改善脂质代谢,调节胆汁酸代谢等有关。Objective To investigate the effect of different doses of Anemarrhenae Rhizoma-Phellodendri Chinensis Cortex herb pair(ZB)on hyperlipidemic rats induced by high-fat diet and its possible mechanism.Methods Sixty male SD rats were randomly divided into 10 normal groups and 50 model groups.The normal group was fed with normal feed,and the model groups were fed with high-fat diet for 24 weeks;on the 21st week of modeling,the rats were randomly divided into the model group,the Anemarrhenae Rhizoma-Phellodendri Chinensis Cortex herb pair for high,medium and low dose groups(3.00,1.00,0.33g·kg^(-1)·d^(-1))the pioglitazone hydrochloride positive drug group(5.25mg·kg^(-1)·d^(-1)),10 rats in each group,the normal group and the model group were given an equal volume of distilled water for 22 days,and the rats were treated at the weekend of 24 weeks.Serum and liver tissues were taken to measure serum triglyceride(TG)and cholesterol(CHO)and total bile acid(TBA)levels were changed,and pathological changes of liver tissues(HE staining)were observed.Results Compared with the normal group,the serum levels of TG and CHO in the model group were significantly increased(P<0.01),and the serum TBA levels were also significantly increased(P<0.05);compared with the model group,the high dose group of Anemarrhenae Rhizoma-Phellodendri Chinensis Cortex herb pair The levels of TG and TBA in the middle-dose group of Anemarrhenae Rhizoma-Phellodendri Chinensis Cortex herb pair significantly decreased(P<0.01),the level of TG in the pioglitazone hydrochloride group decreased significantly(P<0.05),and the level of TBA in the low-dose group of Anemarrhenae Rhizoma-Phellodendri Chinensis Cortex herb pair and pioglitazone hydrochloride group decreased significantly(P<0.01),histopathology showed that Anemarrhenae Rhizoma-Phellodendri Chinensis Cortex herb pair significantly reduced the degree of hepatic cell steatosis,balloon-like degeneration and inflammation of rats in each dose group.Conclusion Anemarrhenae Rhizoma-Phellodendri Chinen-sis Corte
