摘  要:目的通过数据非依赖采集(DIA)技术筛选荔枝核总黄酮(TFL)对大鼠肝星形细胞T6细胞株(HSC-T6)的差异表达蛋白,并进行对差异蛋白进行鉴定和生物信息学分析。方法大鼠随机分为模型对照组和TFL实验组,分别给予相应干预后取大鼠的血清。药物和含药血清干预肝星形细胞后,提取细胞蛋白,通过DDA谱图库结合DIA技术的方法检测模型对照组和TFL实验组的差异表达蛋白,并对差异表达蛋白进行生物信息学分析。结果共鉴定出差异蛋白177个,其中显著下调蛋白共93个,上调蛋白共84个,主要参与了众多与细胞转录、复制、增殖相关的信号通路。结论TFL可能通过调控多个蛋白,参与翻译后修饰、转录、重组和信号传导等多种复杂的信号通路,从而促进肝星状细胞凋亡,达到抗肝纤维化的作用。Objective Through data independent acquision(DIA)technology,the differential expression protein of TFL on rat hepatic stellate cell strain T6(HSC-T6)was screened,and the differential protein was identified and bioinformatics analyzed.Methods Rats were randomly divided into two groups:model control group and TFL experimental group.After intervention of drugs and serum containing drugs on hepatic stellate cells,cell protein was extracted.The differential expression proteins of the model control group and TFL experimental group were detected by the method of DDA spectrum library and DIA technology.Finally,bioinformatics was used to analyze the differentially expressed proteins.Results A total of 177 differential proteins were identified,including 93 down-regulated proteins and 84 up-regulated proteins.They are mainly involved in many signaling pathways related to cell transcription,replication and proliferation.Conclusion TFL may regulate many proteins and participate in many complex signal pathways such as post-translational modification,transcription,recombination and signal transduction,so as to promote the apoptosis of hepatic stellate cells and achieve the effect of anti-hepatic fibrosis.
