摘  要:目的基于网络药理学探讨黄芩治疗RSV的潜在有效成分及作用机制。方法基于TCMSP数据库获取黄芩化学成分及其对应靶点,基于GeneCards数据库获取呼吸道合胞病毒(Respiratory syncytial virus,RSV)肺炎相关靶点,利用Venny 2.1绘制黄芩靶点与RSV肺炎疾病靶点的韦恩图,利用Cytoscape 3.6.1软件构建中药-靶点网络及应用STRING平台构建蛋白相互作用网络(PPI),最后,应用R语言软件对关键靶点进行GO富集分析和KEGG通路富集分析。结果共获得36个化合物,有效成分中度值较高的为新黄芩素、二甲氧基黄酮、5,7,4′-三羟基-8-甲氧基黄烷酮等。PPI网络中核心基因为半胱天冬酶3、Jun原癌基因、丝氨酸/苏氨酸激酶1、血管内皮生长因子A等;GO富集分析显示BH域结合、死亡域结合为显著性最高的通路,其次是细胞因子结合受体等;KEGG富集通路分析晚期糖基化终末产物及其受体信号通路显著性最高,其次是乙型肝炎信号通路、IL-17信号通路、TNF信号通路等。结论文中系统地探讨了黄芩治疗RSV肺炎潜在的有效成分及作用机制,为今后进一步实验验证及临床新药研发提供依据。Objective To explore the potential effective ingredients and mechanism of ScutellariabaicalensisGeorgi in the treatment of RSV based on network pharmacology.Methods Obtaining Scutellariabaicalensis chemical composition and its corresponding target based on TCMSP database.Acquire the targets of respiratory syncytial virus(RSV)pneumonia based on Gene Cards database.Using Venny2.1 to draw the Venn diagram of Scutellaria target and RSV pneumonia disease target.UseCytoscape 3.6.1 software to construct traditional Chinese medicine-target network and STRING platform to construct protein interaction network(PPI).Finally,use R language software to perform GO enrichment analysis and KEGG pathway enrichment analysis on key targets.Results A total of 36 compounds were obtained,among which the active ingredients with higher moderate values were neoscutellarin,dimethoxyflavonoids,5,7,4′-trihydroxy-8-methoxyflavanone and so on.The core genes in the PPI network are caspase 3,Jun proto-oncogene,serine/threonine kinase 1,vascular endothelial growth factor A,etc.GO enrichment analysis shows that BH domain binding and death domain binding are the most significant Pathway,followed by cytokine binding receptors,etc.;KEGG enrichment pathway analysis of advanced glycation end products and their receptor signaling pathways is the most significant,followed by hepatitis B signaling pathway,IL-17 signaling pathway,and TNF signaling pathway etc.Conclusion This study systematically exploredthe potential effective components and mechanism of action of Scutellariabaicalensis in the treatment of RSV pneumonia,and provided a basis for further experimental verification and clinical new drug development in the future.
