摘  要:为寻找一条适合工业化生产高良姜素的路线,本研究采用乙醇提取法、D101型大孔树脂柱色谱法和结晶法相结合的方法,并对各步骤的工艺参数进行了详细优化,成功得到高纯度高良姜素。首先用8倍量的85%乙醇水溶液(体积比)提取高良姜,回流3次,每次2h,得到粗提物,将提取液以1 BV(Bed Volume,树脂床体积)/h的流速在D101大孔树脂柱上负载15 BVs,再用85%乙醇溶液洗脱除去杂质,得到高良姜素粗品,最后以60%乙醇进行重结晶处理,即得纯度达95%高良姜素,经中试放大试验证明该工艺操作简便,稳定可行,为高良姜素工业化生产提供实验依据。In this study,ethanol extraction method,D101 macroporous resin column chromatography and crystallization method were used to optimize the process parameters of each step,and galangin was successfully extracted and purified.Alpinia officinarum was extracted with 8 times of 85%ethanol aqueous solution(volume ratio)and refluxed for 3 times,2 hours each time.The crude extract was obtained by loading 15bvs on D101 macroporous resin column at the flow rate of 1BV/h.The impurities were removed by elution with 85%ethanol solution.The purity of galangin was 95%after recrystallization with 60%ethanol.The pilot scale-up test showed that the process was simple,stable and feasible,which provided experimental basis for industrial production of galangin.
