摘  要:目的:研究平喘颗粒对气道高反应大鼠气道重塑过程中肺组织中Ⅰ型胶原(COL-Ⅰ)、Ⅲ型胶原(COL-Ⅲ)、转化生长因子-β1(TGFβ1)蛋白表达的影响。方法:将48只SPF级健康6周龄Wistar大鼠分为空白组、模型组、治疗组,每组16只。空白组用生理盐水代替进行致敏和激发,其余两组在实验开始后1 d、14 d和21 d,给予0.2 mL抗原液皮下注射致敏,同日用含有高温杀死的6×10^(9)/mL百日咳杆菌菌苗1 mL腹腔注射,并于第28~34天将其放入自制玻璃箱中,给予1%OVA的生理盐水溶液持续雾化30 min,1次/d,连续定时激发5 d,后每隔日激发1次,共激发6周,造成气道高反应大鼠模型。治疗组在末次致敏后第4天开始给予中药(平喘颗粒)灌胃,药物灌胃剂量为1 mL/100 g,空白组和模型组用相应剂量生理盐水(1 mL/100 g)灌胃。有激发阶段时,则在激发前1 h完成药物灌胃。各组大鼠于末次激发24 h后处死,取肺组织进行HE和Mallory染色,观察各组大鼠肺组织病理HE染色、Mallory染色及免疫组化和Western blotting检测肺组织中COL-Ⅰ、COL-Ⅲ、TGFβ1的含量。结果:HE和Mallory染色显示,平喘颗粒能减轻气道高反应大鼠的肺组织形态学改变和胶原沉积。免疫组化法检测结果显示,与空白组相比,治疗组、模型组COL-Ⅰ、CⅠOL-Ⅲ表达均显著升高(均P<0.01);与模型组相比,治疗组COL-Ⅰ、COL-Ⅲ表达均明显降低(均P<0.05)。Western blotting检测结果显示,与空白组相比,治疗组、模型组COL-Ⅰ、COL-Ⅲ、TGFβ1表达均显著升高(P<0.01);与模型组相比,治疗组COL-、COL-Ⅲ、TGFβ1表达明显降低(均P<0.05)。结论:平喘颗粒可以抑制气道高反应大鼠的气道重塑,抑制肺成纤维细胞激活,使TGFβ1的表达降低,减少了ColⅠ和ColⅢ的表达,减少了基底膜增厚和胶原沉积,发挥了抗纤维化的作用。Objective:To study the effect of Pingchuan granules on the expression of type I collagen(COL-I),typeⅢcollagen(COL-Ⅲ)and transforming growth factor-β1(TGFβ1)protein expressionin lung tissues during airway remodeling in hyperresponsive rats.Methods:48 SPF healthy 6-week-old Wistar rats were divided into blank group,model group,and treatment group,with 16 rats in each group.The blank group was sensitized and challenged with normal saline.The other two groups were injected with 0.2 mL antigen solution subcutaneously into the inner limbs and abdominal cavity of the rats at 1 d,14 d and 21 d after the start of the experiment.The same daily use contained high temperature.The killed 6×10^(9)/mL B.pertussis vaccine was injected intraperitoneally with 1 mL,and placed in a 30 cm×30 cm×20 cm self-made glass box connected to the small animal nebulizer on the 28th to 34th day.Give 1%OVA physiological saline solution for 30 minutes,once a day,continuous timed excitation for 5 days,and then once every other day,a total of 6 weeks of excitation,resulting in airway hyperresponsive rat model.The rats in the treatment group were given Pingchuan granules by gavage at the dose of 1 mL/100 g on the 4th day after the last sensitization.The blank group and the model group rats were given equal volume of normal saline.When there was a challenge phase,the drug was administered to the stomach 1 h before the challenge.Rats in each group were sacrificed 24 h after the last challenge.The lung tissues were taken for HE and Mallory staining.The immunohistochemistry and Western blot were used to detect the COL-I,COL-Ⅲand TGFβ1 protein levels in lung tissues.Results:HE and Mallory staining showed that Pingchuan granules could reduce the morphological changes and collagen deposition of lung tissue in rats with airway hyperresponsiveness.Immunohistochemical results showed that compared with the blank group,the expression of COL-I and COL-Ⅲin the treatment group and the model group were significantly increased(P<0.01);compared with the
