摘　　要：目的研究Sirt1和连接蛋白在银屑病皮损中的表达,以及Sirt1的异常表达与活化对体外培养的人永生化角质形成细胞株(HaCaT细胞)连接蛋白表达水平的影响。方法①利用免疫组化技术检测银屑病皮损中Sirt1、紧密连接蛋白(ZO-1、occludin)、黏着连接蛋白(E-cadherin)的表达与定位。②体外培养皮肤HaCaT细胞,利用Sirt1的特异性激活剂(白藜芦醇)和抑制剂(烟酰胺)诱导Sirt1异常表达,经反转录聚合酶链反应(RT-PCR)检测Sirt1激活剂、抑制剂对Sirt1基因表达水平的影响:利用蛋白质印迹试验(Western blotting)检测相同条件下Sirt1、紧密连接蛋白、黏着连接蛋白的蛋白表达水平。结果①与正常人皮肤组织相比,Sirt1在银屑病皮损中表达水平下降,连接蛋白ZO-1、occludin、E-cadherin表达水平均显著升高。②与对照组相比,白藜芦醇可显著增加HaCaT细胞Sirt1的基因和蛋白表达水平,Sirt1的表达与活化明显降低了HaCaT细胞连接蛋白的表达水平。结论Sirt1可能具有下调银屑病角质形成细胞间连接蛋白表达的功能。Objective To investigate the expression of Sirt1 and connexin in psoriatic lesions, and the effect of abnormal expression and activation of Sirt1 on the expression level of connexin in cultured HaCaT keratinocytes in vitro. Methods The expression and localization of Sirt1, ZO-1, occludin and E-cadherin in psoriatic lesions were detected by immunohistochemistry. Sirt1 specific activator(resveratrol) and inhibitor(nicotinamide) were used to induce abnormal expression of Sirt1 in cultured skin keratinocyte line HaCaT in vitro. The effect of Sirt1 activator and inhibitor on the expression of Sirt1 gene was detected by RT-PCR, and the expression levels of Sirt1, tight junction protein and adhesion junction protein were detected by Western blotting under the same conditions. Results To be compared with the normal skin tissue, the expression of Sirt1 in psoriatic lesions decreased, while the expression levels of connexin ZO-1, occludin and E-cadherin increased significantly. To be compared with the control group, in HaCaT cells, resveratrol significantly increased the expression of Sirt1 gene and protein, and the expression and activation of Sirt1 significantly decreased the expression of connexin. Conclusion Sirt1 may down-regulate the expression of connexin in keratinocytes of psoriasis.
