摘 要:目的:优化沙棘果多糖提取工艺。方法:在单因素试验中,选择超声波功率、提取温度和提取时间为考察因素;应用Box-behnken中心组合设计试验和响应面分析法,考察料液比、提取时间、提取功率、提取温度对沙棘果多糖提取率和多糖含量的影响。结果:单因素试验中,最优的提取方案是料液比为1:30,提取时间为45min,提取功率为100W,提取温度为60℃,提取率最高为3.95%,多糖含量为34.90%。响应面分析法试验中,最优的提取方案是料液比为1:30.88,提取时间为45.29min,提取功率为90W,提取温度为61.48℃,提取率最高为4.02%,多糖含量为35.25%。结论:经响应面分析法优化的提取方案具有可行性。Objective:To optimize the extraction technology of polysaccharide from seabuckthorn fruit.Methods:The feasibility of Box-Behnken central composite design test,response surface analysis and single factor analysis were compared to optimize the extraction scheme.The single factor analysis selected ultrasonic power,extraction temperature and extraction time as the investigation factors.The effects of solid-liquid ratio,extraction time,extraction power and extraction temperature on the extraction yield and polysaccharide content of seabuckthorn fruit were investigated.Results:In the single factor experiment,the optimal extraction scheme was solid-liquid ratio of 1:30,extraction time of 45 min,extraction power of 100 W,extraction temperature of 60℃,the highest extraction rate of 3.95%,polysaccharide content of 34.90%.In response surface analysis experiment,the optimal extraction scheme was as follows:solid-liquid ratio of 1:30.88,extraction time of 45.29 min,extraction power of 90 W,extraction temperature of 61.48℃,extraction rate of 4.02%,polysaccharide content of 35.25%.Conclusion:The extraction scheme optimized by response surface analysis is feasible.
