华蟾毒它灵降低ARF6蛋白的表达抑制人卵巢癌细胞SKOV3的增殖

摘　　要：目的探究二磷酸腺苷核糖基化因子6(ARF6)蛋白在人卵巢癌中的表达情况及对患者预后的影响,并初步探讨华蟾毒它灵(CINO)抑制人卵巢癌细胞SKOV3增殖的可能机制。方法免疫组化染色检测人正常卵巢组织及浆液性卵巢癌组织内ARF6蛋白的表达情况,采用Kaplan-Meier生存曲线分析ARF6蛋白表达水平对患者预后的影响。选取人卵巢癌细胞SKOV3,以0、0.5、1.0、2.0μmol/L的CINO分别作用于细胞,采用蛋白质免疫印迹法检测药物处理后的SKOV3中ARF6蛋白的表达。结果浆液性卵巢癌组织内ARF6蛋白的阳性表达率高于正常卵巢组织,ARF6蛋白阳性表达的患者3年总生存率比阴性表达患者低,差异有统计学意义(P<0.05);0.5、1.0、2.0μmol/L的CINO均能抑制卵巢癌SKOV3细胞中ARF6蛋白表达。结论ARF6蛋白在浆液性卵巢癌组织中高表达,且与患者不良预后有关,ARF6蛋白可能是浆液性卵巢癌患者的早期诊断及预后评价的有效分子标志物。CINO抑制人卵巢癌细胞SKOV3的体外增殖能力可能与其下调ARF6蛋白的表达有关。Objective To explore the expression of ADP-ribosylation factor 6(ARF6)protein in ovarian cancer and its influence on the prognosis of patients.And to explore the mechanism of Cinobufotalin inhibiting the proliferation of SKOV3 cell.Methods The expression of ARF6 protein in human normal ovarian tissue and human serous ovarian cancer tissue was detected by immunohistochemical staining.The Kaplan-Meier survival curve method was used to analyze the effect of ARF6 protein on the prognosis of patients.Cell cultures were treated with Cinobufotalin(0,0.5,1.0 and 2.0μmol/L)for 48 h.Expression of ARF6 protein was evaluated by Western Blot analysis.Results The expression of ARF6 protein in serous ovarian cancer tissue was higher than that in normal ovarian tissue,while the 3-year overall survival rate of patients with positive expression of ARF6 protein was lower than the negative expression of ARF6 protein(P<0.05).0.5,1.0,2.0μmol/L Cinobufotalin could inhibit the expression of protein ARF6 protein in ovarian cancer SKOV3 cells.Conclusion The high expression of ARF6 protein in serous ovarian cancer related to patients′poor prognosis.ARF6 protein might be an effective molecular marker for early diagnosis and prognostic evaluation of patients with serous ovarian cancer.The ability of Cinobufotalin to inhibit the proliferation of ovarian cancer SKOV3 cells in vitro might relate to its down-regulation of the expression of ARF6 protein.
