摘  要:目的观察电针预处理"内关"、"足三里"、"关元"穴,对心肌缺血再灌注损伤(MIRI)大鼠心脏组织损伤的改善作用,以及对细胞自噬标志物微管相关蛋白1轻链3Ⅱ(LC3Ⅱ)和细胞凋亡标志物半胱天冬酶-3(Caspase-3)的影响。方法将雄性SPF级Wistar大鼠40只随机分为正常组、假手术组、模型组和电针组各10只。结扎冠状动脉前降支20min再灌注40min后检测血清肌钙蛋白(cTnI)、血清乳酸脱氢酶(LDH)、肌酸激酶(CK)含量、Caspase-3的mRNA表达水平、LC3Ⅱ的蛋白表达水平及心肌细胞凋亡指数(AI)。结果与正常组相比,假手术组cTnI、LDH、CK、Caspase-3 mRNA、LC3Ⅱ蛋白含量及AI等指标均无显著性变化(均P>0.05);与正常组、假手术组相比模型组、电针组血清cTnI、LDH和CK含量均升高(均P<0.05),且电针组含量低于模型组(P<0.05);与正常组、假手术组相比,模型组、电针组AI均显著升高(均P<0.05),且与模型组相比,电针组显著降低(P<0.05);与正常组、假手术组相比,模型组、电针组Caspase-3表达均显著升高(均P<0.05),且与模型组相比,电针组显著降低(P<0.05)与正常组、假手术组相比模型组、电针组LC3Ⅱ表达显著升高(均P<0.05)且与模型组相比,电针组明显下降(P<0.05)。结论电针预处理能有效抑制细胞自噬、降低细胞凋亡水平进而起到保护心脏的作用。Objective To observe the protective effect of electroacupuncture(EA) al "Neiguan", "Zusanli" and "Guanyuan" on myocardial cells of MIRI rats and its effect on LC 3 II and Caspase-3.Methods Forty SPF male Wistar rats were randomly divided into normal control group,sham operation group,model group and EA pretreatment group,with 10 rats in each.The model was established by ligating the anterior descending branch of coronary(artery for 20 min and reperfusion for 40 min).The levels of serum cardiac troponin I(cTnI),serum lactic dehydrogenase(LDH),Caspase-3 mRNA,LC3Ⅱ and apotosis index(AI) were detected.Results There was no significant difference in the above indexes between sham operation group and normal control group(all P>0.05);compared with normal group and sham operation group,the serum cTnI,LDH,CK levels in model group and EA pretreatment group were all increased(all P<0.05),and were all lower in EA pretreatment group than model group(P<0.05);compared with normal control group and sham operation group,the AI in model group and EA pretreatment group were significantly increased(P<0.05),and compared with normal group and shan operation group the AI in EA pretreatment group and model group was significantly decreased(P<0.05);compared with normal control group and sham operation group,the expression of Caspase-3 in model group and EA pretreatment group were significantly increased(all P<0.05),and compared with model group,the expression of Caspase-3 in EA pretreatment group was significantly decreased(P<0.05);compared with normal control group and sham operation group,the level of LC3Ⅱ in model group and EA pretreatment group increased(all P<0.05);compared with model group,the expression of LC3Ⅱ in EA pretreatment group decreased(P<0.05).Conclusion EA pretreatment can effectively inhibit autophagy,reduce the level of apoptosis,and then protect the heart.
