摘 要:目的构建半夏抗癌的成分-靶点-通路"网络,探究半夏可能抗癌活性成分及其潜在作用机制。方法通过文献和TCMSP数据库查找半夏化学成分采用类药性(DL)以及口服生物利用度(OB)评估筛选活性成分;根据反向药效团匹配方法,利用Drugbank、Pharmmapper数据库筛选半夏活性成分潜在靶点基因,通过GeneCards和MalaCards数据库得到癌症相关基因靶点;将活性成分的潜在靶点基因与癌症相关基因进行比对,得到半夏抗癌的潜在靶点基因,使用KOBAS3.0数据库进行KEGG通路注释和GO富集采用Cytoscape3.6.1软件构建半夏抗癌的成分-靶点-通路"网络。结果半夏可能通过豆甾醇、黄芩苷等10个活性成分,作用于包括EGFR、AKT1 等20个基因靶点从而调节Pathways in cancer、Proteoglycans in cancer、Endocrine resistance等32条信号通路共同起到抗癌的效果。结论构建的"成分-靶点-通路"网络图揭示了半夏通过多成分、多靶点、多通路的形式发挥抗癌作用的特点,为进一步阐释半夏抗癌作用机制及其开发利用提供了科学依据。Objective To construct a " components-targets-pathways" network for anti-cancer in Pinellia Rhizoma,and to explore the possible anti-cancer active components of Pinellia Rhizoma and its potential mechanism.Methods The chemical components of Pinellia Rhizoma were searched by literatures and TCMSP database.The active components were screened by DrugLikeness(DL) and Oral Bioactivity(OB).According to the reverse pharmacophore matching method,using Drugbank,Pharmmappe database screen the potential target genes of Pinellia Rhizoma active components.And obtain cancer-related gene targets were obtained through GeneCards and MalaCards databases;comparing potential target genes of active components with cancer-related genes to obtain potential target genes for Pinellia Rhizoma anti-cancer;the KOBAS3.0 database was used for KEGG pathway annotation and GO enrichment;and Cytoscape 3.6.1 software was used to construct the "components-targetspathways" network of Pinellia Rhizoma anti-cancer.Results Pinellia Rhizoma could regulate pathways in cancer, proteoglycans in cancer,endocrine resistance and other 32 signaling pathways through 10 active components including sterol and baicalin,and act on 20 gene targets including EGFR and AKT1 to cure cancer.Conclusion The "components-targets-pathways" network reveals the characteristics of Pinellia Rhizoma exerting anti-cancer effect through multi-components,multi-targets and multipathways,further explaining the mechanism of anti-cancer effect of Pinellia Rhizoma,which provides a scientific basis for its development and utilization.
