摘  要:目的研究原花青素-明胶复合物的形成机制,制备川芎和芎附挥发油微囊,考察以原花青素交联明胶用于制备载药微囊的可行性。方法对原花青素-明胶复合物进行傅里叶转换红外光谱(FTIR)和差示扫描量热(DSC)分析,以明胶和阿拉伯胶为囊材,原花青素为交联剂,复凝聚法制备芎附微囊,考察微囊中指标成分的体外释放行为。结果FTIR结果表明原花青素可与明胶形成分子间氢键,DSC结果显示交联后的明胶热稳定性增强,最优工艺制得的芎附微囊外观圆整,平均粒径(79.4±5.0)μm,具有明显的缓释作用。结论原花青素可与明胶形成氢键,具有缓释特征,芎附微囊的成功制备证明原花青素交联明胶切实可行。Objectives To study the formation mechanism of proanthocyanidin-gelatin complex,prepare chuanxiong and xiongfu volatile oil microcapsules,and investigate the feasibility of using proanthocyanidin cross-linked gelatin to prepare drug-loaded microcapsules Methods Proanthocyanidin-gelatin complexes were analyzed by FTIR and DSC.Complex coacervation method was applied to prepare the microcapsules using gelatin and gum acacia as the shell material and proanthocyanidins as the cross-linker.The release of the active ingredients from the microcapsules was determined using a dialysis method.Results FTIR results showed that proanthocyanidin played a role in forming intermolecular hydrogen bond with gelatin.Results of DSC demonstrated that the thermostability of gelatin crosslinked by proanthocyanidin was enhanced.The appearance of microcapsules in optimal preparation conditions was smooth with an average particle size of(79.4±5.0)μm.The microcapsules showed sustained-release behavior compared with Chuanxiong and Cyperi volatile oil emulsion.Conclusions Chuanxiong and Cyperi volatile oil microcapsules with sustained-release behavior prove that the mechanism of cross-linking is intermolecular hydrogen bond formed between proanthocyanidin and gelatin.
