摘 要:目的观察野菊花提取物对慢性支气管炎大鼠肺组织病理的影响,并探讨其对肺组织转化生长因子-β1(TGFβ1)/信号传导蛋白Smad3通路的调控机制。方法取60只成年健康SD大鼠,除正常组外均建模,低、中、高浓度组予以0.7、1.4、2.8 g/kg野菊花提取物灌胃,每天1次;阳性组予以50 mg/kg N-乙酰半胱氨酸灌胃,每天1次。观察干预前后血清TGF-β1、肿瘤坏死因子-α(TNF-α)、白细胞介素-6(IL-6)水平变化,肺组织病理改变,肺组织TGF-β1、Smad3 mRNA及蛋白表达。结果干预前,模型组血清TGF-β1、TNF-α、IL-6较正常组升高(P<0.01)。干预后,模型组依然升高(P<0.01),野菊花提取物组和阳性组均较干预前水平降低(P<0.05),且低于模型组(P<0.01);正常组肺组织无明显改变,模型组呈病理改变,野菊花提取物组和阳性组均较模型组减轻,且高浓度组与正常组最为接近;各组病变评分比较,模型组均高于正常组(P<0.01),野菊花提取物组和阳性组均低于模型组(P<0.01);各组肺组织TGF-β1、Smad3 mRNA及蛋白表达比较,模型组均高于正常组(P<0.01),野菊花提取物组和阳性组均低于模型组(P<0.01)。结论野菊花提取物可减轻慢性支气管炎大鼠的炎症反应和肺组织病理变化,推测与下调TGF-β1、Smad3 mRNA及蛋白有关。AIM To observe the effects of Chrysanthemum indicum L.extract on lung histopathology in rats with chronic bronchitis,and to explore its regulatory mechanism on transforming growth factor-β1(TGF-β1)/signal transduction protein Smad3 pathway in lung tissue.METHODS Sixty healthy adult SD rats were induced into models,except for those of the normal group.Once daily gavage of 0.7,1.4 and 2.8 g/kg extracts of C.indicum were given to the low-dose,medium-dose and high-dose C.indicum groups,and 50 mg/kg Nacetylcysteine once a day was given to the positive group.The changes of levels of serum TGF-β1,tumor necrosis factor-α(TNF-α)and interleukin-6(IL-6),lung tissue pathology,TGF-β1,Smad3 mRNA and protein expressions were observed.RESULTS In terms of TGF-β1,TNF-αand IL-6 levels,the model group shared increased levels than the normal group before treatment(P<0.01)and those after treatment(P<0.01);all C.indicum groups and positive group displayed decreased post-treatment levels than those prior to the treatment(P<0.05)and the model group(P<0.01).Obvious pathological changes in lung tissues was not observed in the normal group,but was found in the model group,whose deterioration was not surpassed by the positive group and C.indicum groups,and the high concentration group shared a morphological image closest to the normal group.When assessed with the pathological scores,the expressions of TGF-β1,Smad3 mRNA and proteins in lung tissues,the model group had higher scores and levels than that of the normal group(P<0.01),and the positive group and C.indicum groups had even lower scores and levels than that of the model group(P<0.01).CONCLUSION C.indicum extract can alleviate inflammatory reaction and pathological changes of lung tissue of rats with chronic bronchitis,presumably in relation with down-regulation of TGF-β1,Smad3 mRNA and protein.
