摘 要:目的:探究二甲双胍(metformin,MET)对原代皮层神经元轴突生长的效应,以寻找安全有效的促进神经元再生的药物,用于恢复中枢神经系统损伤。方法:原代培养大鼠胚胎18 d(E18)皮层神经元,加入不同浓度MET处理,采用免疫荧光观察神经元轴突生长状况,细胞计数试剂盒8(cell counting kit-8,CCK-8)检测细胞活力变化,利用硫酸软骨素蛋白多糖(chondroitin sulfate proteoglycans,CSPGs)构建神经元轴突生长抑制模型,免疫荧光观察MET是否可以缓解CSPGs的抑制作用。结果:100μmol/L浓度的MET能够有效促进原代皮层神经元轴突的生长和再生;相比于对照组,MET作用48 h能显著提升细胞活力;MET能够缓解CSPGs对神经元轴突生长的抑制作用。结论:MET可有效促进原代皮层神经元轴突生长及损伤轴突的再生,且能有效缓解CSPGs对皮层神经元生长的抑制作用。Objective:To investigate the effect of metformin(MET)on axonal growth and regeneration of primary cortical neurons and find a safe and effective drug to promote the regeneration of neurons,which can be used to recover the damage of central nervous system.Methods:Primary cortical neurons were cultured and treated with different concentrations of MET.The growth and regeneration of axons were observed by immunofluorescence staining and cell viability was measured by cell counting kit-8(CCK-8).Chondroitin sulfate proteoglycans(CSPGs)were used to establish an axonal growth inhibition model to observe whether MET could alleviate the inhibitory effect of CSPGs.Results:The growth and regeneration of primary cortical neuron axons was effectively promoted by MET at the concentration of 100μmol/L.Compared with the control group,MET significantly increased cell viability after 48 h stimulation.MET can alleviate the inhibitory effect of CSPGs on axon growth.Conclusion:MET can effectively promote the axon growth and regeneration of primary cortical neurons and alleviate the inhibitory effect of CSPGs on cortical neuron growth.
