摘  要:目的:观察培元抗癌汤联合替加氟对小鼠H22肝癌细胞生长抑制作用以及对癌相关成纤维细胞(CAFs)调节作用和对细胞因子CXCL1、CXCL2、CXCL8的影响。方法:制备H22肝癌细胞腹水型瘤株悬液,在健康小鼠腋下接种瘤细胞悬液造模,造模成功后随机分为模型组、替加氟组、中西结合组,分别予0.9%的氯化钠溶液、替加氟、培元抗癌汤联合替加氟进行药物干预。观察各组小鼠一般情况,连续用药两周后处死小鼠取瘤组织称重,并计算抑瘤率;Western blot法检测转移瘤中α-SMA、Vimentin、CXCL1、CXCL2、CXCL8蛋白表达水平。结果:干预后,中西结合组较其他两组比较,一般情况较好,小鼠进食、饮水状况良好,体重有所增加,对外部刺激反应敏感,活动无明显异常。替加氟组、中西结合组的瘤体重量明显低于模型组;中西结合组瘤体重量较替加氟组低,差异有统计学意义(P<0.05)。替加氟组、中西结合组小鼠α-SMA、Vimentin、CXCL1、CXCL2、CXCL8蛋白表达均低于模型组,差异有统计学意义(均P<0.05);中西结合组的上述指标均低于替加氟组,差异有统计学意义(均P<0.05)。结论:培元抗癌汤联合替加氟能调节肿瘤微环境中CAFs及细胞因子CXCL1、CXCL2、CXCL8,抑制小鼠H22肝癌细胞生长。Objective:To observe the effects of Peiyuan Kang’ai decoction combined with tegafur on the growth inhibition of mice H22 hepatoma cells,as well as on the regulation of cancer related fibroblasts(CAFs)and the CXCL1,CXCL2,CXCL8 of cytokines.Methods:H22 suspension of ascites type tumor of hepatoma cells was prepared,and the suspension of tumor cells was inoculated under the armpit of healthy mice.After successful modeling,the suspension was randomly divided into model group,tegafluor group,Chinese and western combination group,and the normal saline,tegafluoride,Peyuan Kang’ai decoction and tegafluoride were used for drug intervention respectively.To observe the general condition of each group of mice,the tumor tissue was weighed and the tumor inhibition rate was calculated after two weeks of continuous administration,and the expression level ofα-SMA,Vimentin,CXCL1,CXCL2,CXCL8 protein in metastatic tumor was detected by Western blot method.Results:Compared with the other two groups,the Chinese and western combination group was generally better,the mice were in good condition of eating and drinking water,weight increased,sensitive to external stimuli,and no obvious abnormal activity.The weight of tumor in the tegafluoride group and Chinese and western combination group was significantly lower than that in the model group.The weight of tumor in Chinese and western combination group was lower than that in the tegafur group(all P<0.05).The expression ofα-SMA,Vimentin,CXCL1,CXCL2,CXCL8 protein in the tegafur group and Chinese and western combination group were lower than those in the model group,difference statistically significant(all P<0.05).The above indexes in the Chinese and western combination group were lower than those in the tegafur group,difference statistically significant(all P<0.05).Conclusion:Peiyuan Kang’ai decoction combined with tegafur can regulate CAFs and cytokines in tumor microenvironment CXCL1,CXCL2,CXCL8,inhibit the growth of H22 hepatoma cells.
