摘 要:目的探讨赤芍对缺血性脑卒中大鼠的促血管新生作用及分子机制。方法将SD大鼠随机分为假手术组,模型组,给药组低、中、高剂量组,分析大鼠神经功能,检测脑梗死容积,组织病理变化,脑微血管密度,细胞凋亡以及PGC-1α、Nrf2、VEGF、VEGFR-2蛋白表达。结果与模型组比较,赤芍可改善缺血性脑卒中大鼠的神经功能,缩小脑梗死灶,减轻组织病理损伤,减少细胞凋亡,增加脑微血管密度,促进脑组织PGC-1α、Nrf2、VEGF、VEGFR-2蛋白表达(P<0.05)。结论赤芍激活PGC-1α/Nrf2信号通路,诱导VEGF/VEGFR-2高表达,促进血管新生,发挥抗缺血性脑卒中作用。Objective To evaluate the efficacts of paeoniae on angiogenesis and its molecular mechanism in the ischemic stroke rats.Methods SD rats are randomly divided into sham group,model group and group with low,medium,high dose of paeoniae.The neurological function,cerebral infarction volume,histopathological,apoptosis,microvessel density and PGC-1α,Nrf2,VEGF,VEGFR-2 protein expression methods are used to study the efficacies of paeoniae in ischemic brain.Results Paeoniae is administration improved nerve function impairment,reduced cerebral infarction volume,ameliorated the histopathological damage,reduced the number of apoptotic cells,increased microvessel density and increased expression level of neuron PGC-1α,Nrf2,VEGF and VEGFR-2 protein(P<0.05).Conclusion Paeoniae could reduce cerebral ischemia injury by promoting angiogenesis through activating the signaling pathway of PGC-1α/Nrf2,and including the high expression of VEGF/VEGFR-2.
