摘 要:目的建立测定乌蕨中原儿茶酸、原儿茶醛、牡荆苷的定量分析方法。方法采用Dikmatech Diamonsil Plus C18(250 mm×4.6 mm,5μm)色谱柱,以乙腈-0.4%磷酸溶液为流动相进行梯度洗脱,检测波长为260 nm(原儿茶酸、原儿茶醛)、340 nm(牡荆苷),流速为1.0 mL/min,柱温为30℃,进样量为10μL。结果在建立的检测条件下,3个指标性成分分离度良好,原儿茶酸、原儿茶醛、牡荆苷进样量分别在0.05026~0.5026、0.05640~0.5640和0.14304~1.4304μg范围内线性关系良好(r≥0.9998);精密度、稳定性(24 h内)、重复性试验的RSD均低于2.0%(n=6);平均回收率在97.23%~103.36%,RSD均小于5.0%(n=6);且不同产地乌蕨中3种成分含量差异较大。结论建立的方法操作简便、准确、重复性好,可用于乌蕨药材中原儿茶酸、原儿茶醛、牡荆苷含量的测定,为乌蕨的质量控制提供参考。Objective To establish a quantitative method for determination of protocatechuic acid,protocatechuic aldehyde and vitexin in Stenoloma chusanum.Methods HPLC experiment is performed on a Dikmatech Diamonsil Plus C18 column(250 mm×4.6 mm,5.0μm)with mobile phase consisted of acetonitrile-0.4%phosphoric acid solution(gradient elute)at the flow rate of 1.0 mL/min,the detection wavelength is 260 nm(protocatechuic acid,protocatechuic aldehyde)and 340 nm(vitexin).The column temperature is maintained at 30℃,and sample size is 10μL.Results Under this condition,the results show that the separation of the three components is good.Protocatechuic acid,protocatechuic aldehyde and vitexin show good linear relationship within the range of 0.05026~0.5026,0.05640~0.5640 and 0.14304~1.4304μg,respectively(r≥0.9998).RSD of precision,stability tests(within 24 h)and reproducibility are all lower than 2.0%(n=6).The average recoveries of protocatechuic acid,protocatechuic aldehyde and vitexin are among 97.23%~103.36%with RSD below 5.0%(n=6).The contents of three active components in Stenoloma chusanum from different habitats are quite different.Conclusion The method is simple and accurate with good reproducibility.It is suitable for determining the contents of protocatechuic acid,protocatechuic aldehyde and vitexin in Stenoloma chusanum,providing a scientific basis for the comprehensive quality control of Stenoloma chusanum.
