摘 要:通过集成微流控流体操控、反应控制和电子控制3个系统,成功研制了模块化微流控芯片正电子发射计算机断层扫描(positron emission tomography,PET)显像剂合成仪。将不同结构的微流控芯片进行组合,实现富集、控温合成、分离、纯化等功能的模块化,以满足不同PET显像剂的合成制备要求。实验结果表明,用该仪器自动化合成^(18)F-FDG显像剂仅需25 min,放射化学产率为48%~56%(未衰减校正,n>20),放射化学纯度高于98%,单次合成剂量为10~50 mCi,达到临床使用要求。A PET imaging agent synthesizer based on modular microfluidic chips is successfully developed.It integrates a microfluidic fluid handling system,a reaction control system and an electronic control system.Different microfluidic chips are designed to realize the modularization of enrichment,synthesis,separation,purification and other synthesis-related functions.By combining these chips,the synthesis of various PET imaging agents with different synthesis process could be achieved flexibly.A series of factors including microfluidic flow path,control circuit and key liquid handling components are considered comprehensively.Finally,the synthesizer is integrated and miniaturized.The automatic synthesis of^(18)F-FDG with this synthesizer took only 25 minutes,the radiochemical yield was 48%-56%(n>20),and the radiochemical purity was higher than 98%.The single synthetic dose was about 10-50 mCi,which meets the needs of clinical dose.
