摘　　要：目的探讨放疗对青春期前大鼠生殖系统的影响。方法将24只4周龄雌性SD大鼠随机均分为三组,A、B组分别接受3.2 Gy和8.0 Gy单剂量全身放疗,C组作为对照组。1周后处死,测量体重、卵巢和子宫湿重及脏器系数;检测血清雌二醇、抗缪勒氏管激素(AMH)和孕激素水平;HE染色计数各级卵泡数量。结果与C组比较,A、B组大鼠体重、子宫和卵巢湿重较低(P<0.01或P<0.05),B组大鼠子宫脏器系数较低(P<0.01)。A、B组血清AMH水平低于C组(P<0.05)。与C组相比,A组原始卵泡数量和各级卵泡总数下降(P<0.01),原始卵泡比例下降(P<0.05),初级卵泡比例增加(P<0.05)。结论放疗对青春期前大鼠的子宫和卵巢都有损伤,可导致原始卵泡数量减少。Objective To explore the effect of radiotherapy on the reproductive system of prepubertal rats.Methods Twenty-four 4-week-old female SD rats were randomly divided into three groups with 8 rats each.Group A was treated with 3.2 Gy single-dose whole-body radiotherapy, group B was treated with 8.0 Gy single-dose whole-body radiotherapy and group C was taken as the control.The body weight, wet weight of ovary and uterus and organ coefficient were measured after one week.Serum levels of estradiol, anti-Muller’s tube hormone(AMH) and progesterone were detected.HE staining was used to count the number of follicles at all levels.Results Compared with group C,the weight of rats and wet weight of the ovary and uterus were lower in groups of A and B(P<0.01 or P<0.05),and the uterine organ coefficient in group B was lower(P<0.01).Serum level of AMH was lower in groups of A and B than that in group C(P<0.05).Compared with group C,the number of primordial follicles, total number of follicles at all levels and the proportion of primordial follicles were decreased, and the proportion of primary follicles was increased in group A(P<0.01 or P<0.05).Conclusion Radiotherapy damages the uterus and ovaries of prepubertal rats with a reduction of the number of primordial follicles.
