摘  要:目的:研究无比山药丸对腺嘌呤致肾阳虚模型大鼠肾脏病变的改善作用。方法:采用雄性SD大鼠灌胃腺嘌呤100 mg/kg 8 w制备肾阳虚模型,造模同时给予无比山药丸0.5、1、2 g/kg灌胃,连续给药8 w。期间,测定模型大鼠体重、面温、抓力、自主活动、24 h尿量;取血清,检测血清肌酐(Cr)、尿素氮(BUN)、尿酸(UA)水平;称肾脏湿重,HE及Masson染色观察肾脏组织形态学及胶原纤维沉积情况;免疫组化法检测肾脏中TGF-β1蛋白表达。结果:与正常对照组比较,模型对照组体温、面温、抓力、自主活动明显降低(P<0.05或P<0.01);血清Cr、BUN、UA、24 h尿量、肾脏湿重明显增加,肾组织TGF-β1蛋白表达明显上调(P<0.05或P<0.01);与模型对照组比较,无比山药丸组大鼠体重、面温、抓力、自主活动次数明显增加(P<0.05或P<0.01),24 h尿量、血清Cr、BUN和UA水平、肾脏TGF-β1蛋白表达明显下调(P<0.05或P<0.01),肾脏损伤的病变程度减轻。结论:无比山药丸能改善肾阳虚模型大鼠肾功能受损及肾脏病变,并改善肾阳虚证之腰膝酸软、畏寒肢冷、精神萎靡、夜尿频多等。Objective:To study the efficacy of Wubishanyao Pills in improving the renal lesion in Shenyangxu(肾阳虚) model rats induced by adenine. Methods:The male SD rats were treated with intragastric administration of adenine(100 mg/kg) for eight weeks to induce Shenyangxu(肾阳虚) and also with Wubishanyao Pills at 0.5,1 g/kg and 2 g/kg, respectively, for eight weeks. During that period, the body weight, surface temperature, grasping power, autonomous activity and 24-hour urine volume of the modeled rats were measured, followed by the detection of serum creatinine(Cr), blood urea nitrogen(BUN), uric acid(UA) and the weighing of kidney(wet weight). The renal histomorphology and collagen deposition were observed by HE and Masson staining. The TGF-β1 protein expression in rat kidney was detected by immunohistochemistry. Results: Compared with the normal control group, the model group exhibited significantly reduced body temperature, surface temperature, grasping power and autonomous activity(P<0.05 or P<0.01), elevated serum Cr, BUN, UA, 24-hour urine volume and wet kidney weight, and up-regulated TGF-β1 protein expression(P<0.05 or P<0.01). The body weight, surface temperature, grip strength and frequency of autonomous activities of rats in the Wubishanyao Pill groups were significantly increased as compared with those in the model group(P<0.05 or P<0.01), whereas the 24-hour urine volume, serum Cr, BUN and UA levels, and kidney TGF-β1 expression were significantly lowered(P<0.05 or P<0.01). Besides, the renal injury was relieved. Conclusion: Wubishanyao Pills improve the renal function and pathological changes in model rats with Shenyangxu(肾阳虚) and alleviate such symptoms as soreness and weakness of waist and knee, intolerance of cold, cold limb, spiritual fatigue, and frequent nocturia.
