摘  要:目的:探讨茜草防治心肌梗死的作用,并基于网络药理学研究方法,对其防治心肌梗死的作用机制进行预测,为进一步探究茜草的有效成分及作用机制提供研究基础。方法:尾静脉注射垂体后叶素30 U/kg建立小鼠心肌梗死模型,记录小鼠心电图;使用小鼠血清检测CK、LDH、SOD、MDA、T-AOC的含量;HE染色检测心肌组织病理变化情况;通过TCMSP数据库获取茜草的主要化学成分及其靶点,根据2个ADME筛选中药活性组分;通过Gencards、OMIM、TTD、DRUGBANK数据库获取疾病主要靶点,进行生物过程及通路的信息分析。结果:茜草1、2、4 g/kg可以降低心肌梗死模型小鼠血清LDH、MDA含量(P<0.05),升高CK、SOD活力、T-AOC含量(P<0.05),使升高的ST段降低,改善心肌梗死。茜草防治心肌梗死的核心活性成分为β-谷甾醇、7-羟基-8-甲基-4-乙烯基-9、10-二氢菲-1-羧酸、(2R,3R,4S)-4-(4-羟基-3-甲氧基-苯基)-7-甲氧基-2,3-二羟甲基-四氢化萘-6-ol、木酮,核心靶点有PTGS2、PTGS1、PRKCA、RXRA、NCOA2,其防治心肌梗死的生物学通路主要作用于心肌细胞的肾上腺素信号传导通路、PI3K信号通路和VEGF信号通路。结论:本研究初步揭示了茜草具有防治心肌梗死作用,且与降低氧化应激反应、提高心肌细胞抗氧化能力有关,网络药理学预测β-谷甾醇为茜草防治心肌梗死的主要成分,作用机制与心肌细胞的肾上腺素信号传导通路、PI3K信号通路和VEGF信号通路相关。Objective:To explore the effect of Rubiae Radix et Rhizoma on the myocardial infarction,and to predict its mechanism of action based on network pharmacological,so as to provide a research basis for further research on its effective components and the mechanism of action.Methods:The myocardial infarction rat model was established by injecting 3 U/mL pituitrin into the tail vein,and the ECG of the mice was recorded.The contents of CK,LDH,SOD,MDA and T-AOC in mice were detected by Elisa.HE staining was used to detect the pathological changes of myocardial tissue.The main chemical constituents and targets of Rubiae Radix et Rhizoma were obtained through TCMSP database,and the active components of Traditional Chinese medicine were screened according to 2 ADME methods.The main disease targets were obtained through Gencards,OMIM,TTD and DRUGBANK databases,then the subsequent information analysis of biological processes and pathways was conducted.Results:1、2、4 g/kg Rubiae Radix et Rhizoma reduced the contents of LDH and MDA in myocardial infarction rat serum(P<0.05),increased the CK level,SOD activity and T-AOC content(P<0.05).It also reduced the increased ST,and improved the myocardial infarction.The core active components of Rubiae Radix et Rhizoma in the prevention and treatment of myocardial infarction were beta-sitosterol,7-hydroxy-8-methyl-4-vinyl-9,10-dihydrophenanthrene-1-carboxylic acid,(2 R,3 R,4 S)-4-(4-hydroxy-3-methoxy-phenyl)-7-methoxy-2,3-dimethylol-tetralin-6-ol and Xyloidone.The core targets were PTGS2,PTGS1,PRKCA,RXRA,and NCOA2.The biological pathway of Rubiae Radix et Rhizoma in the prevention and treatment of myocardial infarction were mainly the adrenergic signaling pathway,PI3 K signaling pathway and VEGF signaling pathway in cardiomyocytes.Conclusion:This study preliminarily reveals that Rubiae Radix et Rhizoma has the effect of preventing and treating myocardial infarction,which may be related to reducing the oxidative stress response and improving the antioxidant capacity of myocardial cells.
