摘 要:目的:根据文献分析慢性非细菌性前列腺炎(chronic non-bacterial prostatitis, CNP)动物模型的造模方法及其特点,为合理选择动物模型,提高造模成功率及模型评价指标提供参考。方法:以"前列腺炎"、"模型"为主题词,在中国知网数据库(CNKI)中,不限检索时间,搜索进行CNP动物模型研究的文献,统计所用动物情况、造模方法、造模时间、给药时间及检测指标,进行分析。结果:共筛选得340篇文献,包含化学刺激法、去势+雌激素法、自身免疫法3种造模方法,以SD、Wistar大鼠应用居多。3种方法造模周期一般分别为7 d~14 d、30 d、45 d,给药时间均以28 d~30 d居多。检测指标主要为病理形态学检查、前列腺相关指标及血清生化指标,并以炎症因子、氧化应激因子、疼痛介质为主。结论:化学刺激法、去势+雌激素法、自身免疫法模型分别适用于药效、药效与机制研究,以自身免疫法更接近临床表现。可将病理形态学、表观指标、白细胞数目及卵磷脂小体密度、炎症因子、疼痛程度、尿动力学作为评价指标。增加对于病证结合模型的研究、深入探索取代百白破疫苗(DPT)的造模方法,增加对行为学、尿动力学、表观指标的检测,为未来重要发展方向。Objective: According to the literature, the characteristics of animal models of chronic abacterial prostatitis were analyzed in order to provide references for the reasonable selection of animal models, for improving the successful rate of modeling and the evaluation indexes of models. Methods: With "prostatitis" and "model" set as the subject words, the literatures of CNP(chronic non-bacterial prostatitis) animal model were searched in CNKI database without limited search time. The animal conditions, the establishment methods of model, the time of establishment, the administration time and detection indexes were obtained and statistically analyzed. Results: A total of 340 articles were selected, including the chemical stimulation, castration combined with estrogen method and autoimmune method. SD and Wistar rats were mostly used. The establishment periods of the three methods were 7-14 days, 30 days and 45 days, respectively, and the administration time was mainly 28-30 days. The detection indexes included pathomorphological changes, prostate related indexes and serum biochemical indexes. The inflammatory factors, oxidative stress factors and pain mediators were the main indicators. Conclusion: The models of chemical stimulation, castration combined with estrogen, and autoimmune method are suitable for the study of pharmacodynamics, pharmacodynamics and mechanism of action, respectively. And the model induced by the autoimmune method is closer to the disease in the clinical manifestations. Pathomorphology, apparent index, white blood cell counts and the lecithin body density, inflammatory factors, pain levels and urodynamics can be used as evaluation indexes. In the future, the important future direction is to increase the research of disease syndrome combination model, to explore the establishment method to replace DPT vaccine method, and increase the detection of behavior, urodynamics and apparent indicators.
