摘 要:目的研究超声法提取索骨丹中黄酮及多糖的最佳提取工艺。方法通过单因素实验和正交实验,测定黄酮和多糖的提取率,分析料液比(g·mL^(-1))、超声时间、乙醇体积分数、提取次数对索骨丹中黄酮提取率的影响;料液比、超声时间、提取次数对索骨丹中多糖提取率的影响,优选了索骨丹中黄酮和多糖超声提取的最佳工艺条件。结果索骨丹中黄酮的超声提取最佳工艺条件为:提取3次、料液比1∶20、乙醇体积分数70%、提取时间40min,提取率为9.85%;多糖的最佳提取工艺为:提取3次、料液比1∶40、提取时间40min,提取率为1.69%。结论本实验对索骨丹中黄酮及多糖的超声提取工艺进行了优化,为索骨丹药材的质量控制和有效成分的开发利用提供实验参考。Objective The best extraction methods of flavonoids and polysaccharides from RodgersiaaesculifoliaBatal were studied. Method The extraction rate of flavonoids and polysaccharides using as a selection index, by the single factor and orthogonal experiments,the effect of solid-liquid ratio(g·mL^(-1)), ultrasonic time, ethanol volumetric fraction and extraction times on the extraction rate of total flavonoids in RodgersiaaesculifoliaBatal was analyzed,as well as the effect of solid-liquid ratio(g·mL^(-1)), extraction times and ultrasonic time on the extraction rate of polysaccharides in RodgersiaaesculifoliaBatal,and the optimal process conditions for ultrasonic extraction of flavonoids and polysaccharides were optimized. Result The optimum conditions for ultrasonic extraction of flavonoids from RodgersiaaesculifoliaBatal were as follows: extraction 3 times, ratio of solid-liquid ratio1 ∶ 20, ethanol volumetricconcentration 70%, extraction time 40 minutes, the extraction rate of total flavonoids was 9.85%. The optimal extraction process conditions of polysaccharides were as follows: the extraction 3 times, solid-liquid ratio 1∶40, the extraction time 40 min, the extraction rate of polysaccharide was 1.69%. Conclusion This experiment optimized the ultrasonic extraction method of total flavonoids and polysaccharides in RodgersiaaesculifoliaBatal, and provided experimental reference for the quality control and development and utilization of the active ingredients of RodgersiaaesculifoliaBatal.
