摘  要:目的:比较弱精症不育男性和健康可育男性精子(microRNA,miRNA)的表达差异,并预测miRNA在弱精症发生中的分子机制。方法:分析2018年12月—2019年11月深圳市宝安人民医院(集团)收集的弱精症不育男性(35例)和健康可育男性(36例)精液样本,提取R N A并测定其浓度和纯度,选择了4组实验组样本和3组对照组样本进行后续实验。采用二代测序平台分析miRNA的表达水平,筛选差异表达miRNA。通过miRTarBase数据库预测差异表达miRNA的靶基因,并对miRNA靶基因进行深度信息学分析。结果:与健康可育男性组相比,弱精子症不育男性的精子miRNA表达水平发生显著改变。共有16个miRNA的表达水平改变,差异具有统计学意义(|log2(FoldChange)|>1且FDR<0.01),其中10个miRNA表达下调,6个miRNA表达上调。预测到9744个miRNA靶基因。靶基因KEGG通路分析结果表明miRNA主要参与了肿瘤、内分泌抵抗、细胞衰老、乙型肝炎等通路。结论:弱精子症不育男性和健康可育男性的精子miRNA表达谱有显著差异。这些miRNA靶基因涉及的功能及代谢通路非常广泛,通过多种途径对弱精症的发生起到重要调节作用。Objective To compare the differential expression of sperm microRNA(miRNA)in infertile men with asthenozoospermia and healthy fertile men,and to predict the molecular mechanism of miRNA in the occurrence of asthenozoospermia.Methods Analyze semen samples collected from December 2018 to November 2019 for infertile men with asthenozoospermia(35 cases)and healthy fertile men(36 cases).RNA was extracted and their concentration and purity were determined.Four groups of experimental group samples and three groups of control group samples were selected for subsequent experiments.The second-generation sequencing platform was used to analyze the expression level of miRNA and screen the differentially expressed miRNA.The miRTarBase database was used to predict the target genes of differentially expressed miRNAs,and to conduct in-depth informatics analysis of the target genes of miRNAs.Results Compared with the healthy fertile male group,the sperm miRNA expression level of infertile males with asthenozoospermia sperm syndrome changed significantly.A total of 16 miRNAs had statistically significant changes in their expression levels(|log2(FoldChange)|>1 and FDR<0.01),of which 10 miRNAs were down-regulated and 6 miRNAs were up-regulated.9744 miRNA target genes were predicted.Analysis of the target gene KEGG pathway indicates that miRNA is mainly involved in tumor,endocrine resistance,cell senescence,hepatitis B and other pathways.Conclusion There are significant differences in miRNA expression profiles between infertile men with asthenozoospermia and healthy fertile men.These miRNA target genes are involved in a wide range of functions and metabolic pathways,and play an important role in regulating the occurrence of asthenospermia through various pathways.
