摘 要:目的观察含过氧化氢的低温消毒剂的消毒相关性能、稳定性及毒性,为实际应用提供依据。方法采用载体浸泡杀菌试验、模拟现场消毒试验、稳定性试验、低温试验、金属腐蚀性试验以及动物毒性试验等方法。结果该过氧化氢低温消毒剂含过氧化氢16.2 g/L。该消毒剂原液在(20±1)℃、(-20±1)℃对金黄色葡萄球菌、大肠埃希菌分别作用5 min、30 min杀灭对数值均大于3.00,在(20±1)℃对白色念珠菌作用5 min杀灭对数值大于3.00;经54℃恒温放置14 d后,过氧化氢含量下降率小于10%;在-20℃存放15 h,仍保持液体状态,无析出、无结晶;对碳钢、铝、不锈钢基本无腐蚀作用,对铜有中度腐蚀作用;重金属(以Pb计)含量为<0.05 mg/kg,砷(以As计)含量为<0.01 mg/kg。毒性实验结果表明,急性经口毒性试验LD5o>5000 mg/kg,属实际无毒;一次完整皮肤刺激试验结果属无刺激性;对小鼠骨髓嗜多染红细胞无致微核作用。结论该消毒剂在(-20±1)℃条件下有良好的杀菌效果,稳定性好,低温试验和安全性符合相关要求,可用于低温环境下物体表面和冷链物品外包装表面消毒。Objective To observe the disinfection-related properties, stability and toxicity of a lowtemperature disinfectant containing hydrogen peroxide,and to provide basis for practical applications.Methods The methods of carrier immersion sterilization test, simulated field disinfection test,stability test, low temperature test, metal corrosion test and animal toxicity test were adopted.Results This disinfectant contained 16.2 g/L hydrogen peroxide. The killing log value of the stock solution was greater than 3.00 against Staphylococcus aureus and Escherichia coli at(20 ± 1) ℃and(-20 ± 1) ℃ for 5 and 30 minutes, against Candida albicans at(20 ± 1) ℃ for 5 minutes,respectively. After being placed at a constant temperature of 54 ℃ for 14 days, the reduction rate of hydrogen peroxide content was less than 10%. When stored at-20 ℃ for 15 hours,it maintained the liquid state without precipitation or crystallization. It had no corrosive effect on carbon steel,aluminum or stainless steel, but moderate corrosive effect on copper. The content of heavy metals (calculated as Pb) was <0.05 mg/kg, and the content of arsenic(calculated as As)was <0.01 mg/kg. The toxicity test results showed that the disinfectant was actually non-toxic by acute oral toxicity test with the LD50(median lethal dose) >5 000 mg/kg. The result of the complete skin irritation test was nonirritating. There was no micronucleus effect on bone marrow polychromatic erythrocytes in mice.Conclusion The disinfectant has a good bactericidal effect at(-20±1) ℃ and stability,with the low temperature test and safety meeting the relevant requirements. It can be used to disinfect the surface of objects and the outer packaging of cold chain items in low temperature environments.
