摘 要:目的:探讨烧伤后皮肤成纤维细胞中miR-506-3p表达与Beclin-1的靶向关系及增殖调控作用,为烧伤后瘢痕的治疗提供新靶点。方法:采用实时定量聚合酶链反应(qRT-PCR)检测烧伤组织和热损伤皮肤成纤维细胞中miR-506-3p水平;Targetscan在线预测,双荧光素酶验证miR-506-3p与Beclin-1靶向关系;qRT-PCR和Western blot检测miR-506-3p模拟物或抑制剂转染对Beclin-1表达的影响;qRT-PCR检测热刺激后miR-506-3p模拟物(10nM或30nM)转染成纤维细胞miR-506-3p的表达并采用CCK-8检测转染细胞增殖效率。结果:与正常组织相比,烧伤组织和热损伤成纤维细胞中miR-506-3p的表达下调;miR-506-3p和Beclin-1存在靶向调控作用关系,并且miR-506-3p可负调控Beclin-1表达;miR-506-3p模拟物转染以剂量依赖的方式抑制了热刺激的真皮成纤维细胞中的细胞增殖。结论:miR-506-3p通过靶向抑制Beclin-1表达来调节烧伤后皮肤成纤维细胞的自噬和增殖,有可能成为烧伤愈合后瘢痕治疗的潜在靶标。Objective To explore the targeting relationship between the expression of miR-506-3p and Beclin-1 in heat-damaged skin fibroblasts and its proliferation regulation effect on Beclin-1,in order to provide a new target for the treatment of post-burn scars.Methods We used qRT-PCR to detect miR-506-3p of post-burn tissue and heat-damaged skin fibroblasts,used Targetscan online prediction and dual luciferase to verify the targeting relationship between miR-506-3p and Beclin-1.We used qRT-PCR and Western blot to detect the effect of miR-506-3p mimic or inhibitor transfection on Beclin-1 expression.The expression of miR-506-3p in fibroblasts transfected with miR-506-3p mimic(10nM or 30nM)after thermal stimulation was detected by qRT-PCR and the proliferation efficiency of transfected cells was detected by CCK-8.Results Compared with normal tissues,the expression of miR-506-3p in post-burn tissues and heat-damaged fibroblasts is down-regulated.There is a targeted regulation relationship between miR-506-3p and Beclin-1,and miR-506-3p can negatively regulate the expression of Beclin-1,and miR-506-3p mimic transfection inhibits cell proliferation in heat-stimulated dermal fibroblasts in a dose-dependent manner.Conclusion MiR-506-3p regulates the autophagy and proliferation of post-burn skin fibroblasts by targeting the expression of Beclin-1,and may be a potential target on scar for post-burn healing.
