摘  要:目的研究miR-6819-3p及其IL-17信号通路中靶基因在斑秃病变部位的表达。方法收集2020年6-12月于山西省人民医院皮肤性病科诊疗的斑秃患者病变部位头皮和外科手术所获得的正常头皮组织,将其分为斑秃组和正常组。通过荧光定量PCR检测斑秃组和正常组的miR-6819-3p及其IL-17信号通路中靶基因(CSF3、CXCL8、FOS、FOSB、JUN和NFKBIA)的表达情况,并利用生物信息学分析miR-6819-3p与这些预测靶基因间的调控位点、以及它们在IL-17信号通路中的分布和相互作用关系。结果与正常组相比,斑秃组miR-6819-3p表达量降低(P<0.05),而CSF3、CXCL8、FOS、FOSB、JUN和NFKBIA基因表达量均呈现不同程度的增加(P<0.05)。生物信息学分析显示,CSF3、CXCL8、FOS、FOSB、JUN和NFKBIA是miR-6819-3p的潜在靶基因,且是IL-17发挥作用的下游关键基因。结论miR-6819-3p可以通过对靶基因以及IL-17信号通路的调控在斑秃的发病中发挥作用,提示这些基因可以成为斑秃诊断和治疗中的生物靶标。Objective To investigate the expression of mir-6819-3p and its target genes in IL-17 signaling pathway in alopecia areata lesion sites.Methods The lesion tissues of alopecia areata patients and the normal scalp tissues in the Department of Dermatology,Shanxi Provincial People’s Hospital from June to December in 2020 were collected and divided into alopecia areata group and normal group.The expression levels of miR-6819-3p and its target genes(CSF3,CXCL8,FOS,FOSB,JUN and NFKBIA)in IL-17 signaling pathway in alopecia areata group and normal group were detected by fluorescent qPCR.Bioinformatics analysis was used to identify the regulatory sites between miR-6819-3p and these predicted target genes,as well as their distribution in the IL-17 signaling pathway and their interaction.Results Compared with normal group,the expression of miR-6819-3p was decreased in alopecia areata group(P<0.05),whereas the expression levels of CSF3,CXCL8,FOS,FOSB,JUN and NFKBIA were increased to various degrees(P<0.05).Bioinformatics analysis revealed that CSF3,CXCL8,FOS,FOSB,JUN and NFKBIA were potential target genes of miR-6819-3p,and were also downstream key genes of IL-17.Conclusion The miR-6819-3p might play a role in the pathogenesis of alopecia areata by regulating these target genes and IL-17 signaling pathway,which suggests that these genes could become biological targets in the diagnosis and treatment of alopecia areata.
