摘 要:探讨芦荟多糖对Ⅱ型糖尿病小鼠的降糖作用,并从血糖、血脂、胰岛素、胰高血糖素的变化探讨其可能作用途径。方法为:采用高脂膳食法联合腹腔注射四氧嘧啶复制糖尿病模型,将造模成功的小鼠随机分为模型组,二甲双胍组(阳性对照组),芦荟多糖高、中、低(药物干预组)5组。经灌胃给药14 d后,对小鼠尾椎取血,通过血糖仪测定小鼠血糖,取血清部分通过胰岛素、胰高血糖素试剂盒直接测定胰岛素、胰高血糖素,通过分光光度计法测定小鼠血清中血脂水平(HDL、LDL、TG、TC)含量。结果显示,血糖:与模型组相比,二甲双胍组,芦荟多糖高、中剂量组血糖明显降低(P<0.05),且高剂量组较低剂量组血糖降低更显著(P<0.01),而低剂量组差异不显著(P>0.05);胰岛素:与模型组相比,药物干预组胰岛素水平无差异(P>0.05);血清胰高血糖素:与模型组相比,芦荟多糖高、中剂量组的胰高血糖素明显降低(P<0.05);血脂:与模型组比较,芦荟多糖高、中剂量组的HDL显著升高(P<0.05),芦荟多糖高、中、低剂量组的LDL、TG、TC明显降低(P<0.05)。结论:芦荟多糖可以降低Ⅱ型糖尿病小鼠的血糖和胰高血糖素水平,且能升高Ⅱ型糖尿病小鼠的HDL、降低LDL、TG、TC的含量,提示芦荟多糖有较好的降糖作用,还可有效预防糖尿病心血管病的发生。Objective: Investigating the effect of Aloe Polaccharides(AP) on Type Ⅱ diabetic mice, and exploring the possible pathway from the changes of blood glucose, blood lipid, insulin and glucagon.Methods: Type Ⅱ diabetic mice were induced by high fat diet and intraperitoneal injection with alloxan.Then the mice were randomly divided into five groups: model group, metformin group(Positive Control Group),AP high, middle and low dose group(drug intervention group).After 14 days, the mice’s blood was taken from tail vein to measure their Blood Glucose, Blood Lipid(HDL,LDL,TG,TC),Insulin and Glucagon level.Results: Blood Glucose: Compared with the model group, the blood glucose of the metformin group, AP high and middle group were significantly lower(P<0.05),especially the AP high dose group decreased more significantly than that of the low dose group(P<0.01),but no difference was found in blood blucose between the AP low dose group and the model group(P>0.05).Insulin: Compared with the model group, insulin level of drug intervention group had no difference(P>0.05);Glucagon: Compared with model group, the glucagon of the AP high and middle group was significantly lower(P<0.05);Blood Lipid: Compared with model group, HDL increased significantly in high and medium dose groups(P<0.05),and LDL,TG,TC decreased significantly in drug intervention group(P<0.05).Conclusion: AP could reduce blood glucose and glucagon in Type Ⅱ diabetic mice, and could increase HDL,at the same time decrease the level of LDL,TG and TC in Type Ⅱ diabetic mice.These results suggest that AP has a better hypoglycemic effect, and can effectively prevent the occurrence of diabetic cardiovascular disease.
