摘 要:[目的]稀有冷诱导2(RCI2)基因,又称为质膜蛋白3(PMP3)基因,编码一类小分子量的跨膜蛋白质,在植物生长发育和非生物胁迫应答中发挥重要调控作用。本文旨在研究棉花中RCI2基因的特征与表达模式,为进一步了解该基因在棉花中的功能提供理论支持。[方法]本文通过生物信息学方法,对陆地棉、海岛棉、亚洲棉和雷蒙德氏棉4个棉种中RCI2家族基因特征、亚细胞定位、系统进化、启动子顺式元件和非生物胁迫应答模式进行系统分析。[结果]分别从陆地棉、海岛棉、亚洲棉和雷蒙德氏棉基因组中分别筛选到12、12、8、8个RCI2基因,该基因编码的蛋白质仅含有PMP3(PF01679)一个高度保守的结构域。通过进化分析,将棉花中RCI2基因分为3类。顺式作用元件分析结果表明,陆地棉RCI2家族基因的启动子中含有较多的激素响应、胁迫响应和生长发育相关的元件,其中激素响应和胁迫响应的元件数目较多。陆地棉中12个RCI2基因在不同组织器官中表达方式不同,在低温、高温、盐和PEG胁迫下的表达模式也不同。[结论]本研究明确了RCI2基因在棉花基因组中的数量和理化性质,并对该家族的进化和表达特征进行了分析,为进一步研究RCI2基因的作用机理提供了参考,为研究棉花应对非生物胁迫的分子机制提供新的方向。[Objective]Rare cold inducible 2(RCI2),also known as plasma membrane protein 3(PMP3),codes a small molecular weight transmembrane protein,which plays an important role in plant development and abiotic stress response.Present study aimed to investigate the characteristics and expression patterns of the RCI2 gene in cotton,and to provide theoretical support for further understanding of the function of this gene in cotton.[Methods]In this study,the characteristics,subcellular localization,phylogeny and abiotic stress response patterns of RCI2 family genes in cotton were systematically analyzed by bioinformatics methods.[Results]Results indicated that a total of 12,12,8 and 8 RCI2 genes were screened out from the genomes of Gossypium hirsutum,Gossypium barbadense,Gossypium arboreum and Gossypium raimondii,respectively.Only one highly conserved domain of PMP3(PF01679)was observed in the protein encoded by RCI2 gene.Through evolutionary analysis,the RCI2 genes were divided into three categories.Cis-element analysis in promotor showed that the RCI2 gene family in G.hirsutum contained more elements related to hormone response,stress response,and growth and development,among which the number of elements related to hormone response and stress response was higher than others.The 12 RCI2 genes in different tissues and organs of different cotton displayed different expression patterns,and their expression patterns were also different under low temperature,high temperature,salt and PEG stress.[Conclusion]This study clarified the quantity,physical and chemical properties,evolution and expression characteristics of RCI2 genes in cotton genome.It provided a reference for further study of RCI2 gene and a new direction for cotton to deal with abiotic stress.
