摘 要:目的观察脂肪酸结合蛋白7(FABP7)、Toll样受体4(TLR4)在糖尿病周围神经病变(DPN)小鼠脊髓中的表达。方法采用随机数字表法将20只C57BL/6小鼠分为2组:对照组(Ctrl组)、DPN组,每组10只。采用连续2 d腹腔注射链脲佐菌素(100 mg·kg^(-1))复制DPN小鼠模型,Ctrl组小鼠同方法等剂量注射柠檬酸钠缓冲液;测2组0、2、4、6、8和10周时小鼠体重、随机血糖、热缩足潜伏期(TWL)、机械缩足反应阈(MWT);免疫组化法测定2组足底表皮神经纤维密度(INEFD);Western blot法检测2组脊髓FABP7和TLR4的表达;ELISA法测定2组血浆与脊髓中TNF-α水平。结果与Ctrl组比较,DPN组随机血糖(2、4、6、8和10周)、TWL(4、6、8和10周)明显升高(均P<0.05),体重(2、4、6、8和10周)、MWT(2、4、6、8和10周)、IENFD明显下降(均P<0.05)。与Ctrl组比较,DPN组FABP7、TLR4蛋白表达显著上调(均P<0.05),血浆与脊髓组织中TNF-α水平均升高(均P<0.05)。结论DPN小鼠模型中脊髓组织FABP7、TLR4表达均增高,提示FABP7参与DPN发病,可能与FABP7调控TLR4受体影响炎症因子水平有关。Objective To observe the expression of fatty acid binding protein 7(FABP7)and Toll-like receptor 4(TLR4)in the spinal cord of mice with diabetic peripheral neuropathy(DPN).Methods Twenty C57BL/6 mice were randomly divided into two groups,with 10 mice in each group.In the DPN group,mice were intraperitoneally injected with streptozotocin(100 mg/kg)for consecutive 2 days to induce DPN.In the Ctrl group,mice were given the same dose of sodium citrate buffer.Body weight,random blood glucose,thermal withdrawal latency(TWL)and mechanical withdrawal threshold(MWT)were measured by immunohistochemistry at 2,4,6,8 and 10 weeks.The plantar intraepidermal nerve fiber density(IENFD)and expression of FABP7 and TLR4 in the spinal cord were detected by Western blot.The levels of TNF-αin serum and spinal cord were determined by ELISA.Results Compared with the Ctrl group,the random blood glucose concentration increased at 2,4,6,8 and 10 weeks,the TWL increased at 4,6,8 and 10 weeks,and the body weight and MWT decreased at 2,4,6,8 and 10 weeks in the DPN group(P<0.05).The IENFD in the DPN group was lower than that in the Ctrl group(P<0.05).However,the expression of FABP7 and TLR4 and the levels of TNF-αin serum and spinal cord in the DPN group were higher than those in the Ctrl group(P<0.05).Conclusion The expression of FABP7 and TLR4 increases in the spinal cord of DPN mice,suggesting that FABP7 is associated with DPN through regulating TLR4 receptors to affect the levels of inflammatory factors.
