摘　　要：目的:研究泽泻汤加味方治疗盐敏感性高血压机制。方法:60只Dahl雄性大鼠分为6组,除空白组均给予8%NaCl高盐饲料喂养4周。成模后测大鼠收缩压,体质量、肾重、肾重/体质量,PAS染色、电镜观察肾形态学病理变化,Western blot、RTPCR测肾NADPH氧化酶蛋白和mRNA表达。结果:与模型组比较中药组血压降低,肾重、肾重/体质量低,形态学病理变化小,p22phox、p47phox和NOX4蛋白和mRNA表达降低(P<0.05)。结论:泽泻汤加味方能降血压和保护肾功能,其机制与下调NADPH氧化酶有关。Objective:To study the mechanism of Modified Zexie Decoction in the treatment of salt sensitive hypertension.Methods:60 male Dahl salt sensitive rats were divided into 6 groups.Except the blank group,all rats were fed with 8%NaCl high salt for 4 weeks.The systolic blood pressure,body weight,kidney weight and ratio of kidney to body weight were measured.The morphological and pathological changes of kidney were observed by PAS staining and electron microscope.The protein and mRNA expression of NADPH oxidase were detected by Western blot and RT-PCR.Results:Compared with the model group,the blood pressure of Modified Zexie Decoction groups were decreased,the ratio of kidney weight to body weight were lower,the morphological and pathological changes were small,the protein and mRNA expression of p22phox,p47phox and NOX4 were decreased(P<0.05).Conclusion:Modified Zexie Decoction can reduce the blood pressure and protect renal function,which is related to the down regulation of NADPH oxidase.
