摘  要:目的:观察二陈汤通过增强自噬改善痰湿型多囊卵巢综合征(polycystic ovary syndrome,PCOS)大鼠子宫自噬相关基因7(autophagy related gene,Atg7)、磷脂酰肌醇3激酶(phosphatidylinositol 3 kinase,PI3K)/蛋白激酶B(protein kinase B,Akt)/葡萄糖转运蛋白4(glucose transporter protein 4,Glut4)信号通路的作用,探讨二陈汤改善痰湿型PCOS胰岛素抵抗(insulin resistance,IR)的可能机制。方法:选取动情周期规律SD大鼠42只,运用高脂+来曲唑灌胃法复制痰湿型PCOS大鼠,随机分为对照组10只和模型组32只。造模结束后,周期延长的模型组大鼠随机分为模型组、中药组和二甲双胍组各10只。Elisa法测定大鼠血清激素水平,胰岛素抵抗指数(homeostasis model assessment-Insulin resistance,HOMA-IR),HE染色观察子宫病理组织形态,Western Blot法检测子宫Atg7、PI3K、Akt及Glut4蛋白表达水平。结果:与对照组比较模型组血清性激素水平普遍升高(P<0.05),与模型组比较2组药物治疗组性激素水平降低(P<0.05),与对照组比较模型组HOMA-IR升高(P<0.05),与模型组比较药物治疗组HOMA-IR降低(P<0.05),与对照组比较模型组子宫Atg7、PI3K、Akt及Glut4蛋白水平降低(P<0.05),与模型组比较药物治疗组子宫Atg7、PI3K、Akt及Glut4蛋白水平有一定改善(P<0.05)。结论:二陈汤能够显著抑制PCOS IR水平,其机制可能是通过提高子宫内膜的自噬能力、增强PI3K/Akt/Glut4信号通路实现的。Objective:To investigate the function of Erchen decoction improves the function of Atg7,PI3K/Akt/Glut4 in uterine tissue of rats with PCOS rats with phlegm dampness syndrome by enhancing autophagy.To investigate the possible mechanism of Erchen decoction in improving insulin resistance of PCOS.Methods:Forty-two SD rats with regular estrous period were selected.PCOS rats with phlegm dampness Syndrome were induced by filling the stomach with high in fat and letrozole.Rats were divided into the control group(n=10)and model group(n=32).After modeling,rats with extended estrous period were selected as the model group(n=10),the TCM group(n=10)and the metformin group(n=10).Serum hormone levels and HOMA-IR index were measured by ELISA.Uterine pathological morphology was observed by HE staining,the protein expression of Atg7,PI3K,Akt and Glut4 were detected by Western Blot.Results:Compared with the control group,the serum levels of sex hormones in model group were increased(P<0.05);Compared with the model group,sex hormones in drug treatment-groups were decreased(P<0.05).Compared with the control group,HOMA-IR in model group were increased(P<0.05).Compared with the model group,HOMA-IR in drug treatment groups were decreased(P<0.05).Compared with the control group,the expression of Atg7,PI3K,Akt and Glut4 in model group were decreased(P<0.05).Compared with the model group,the expression of Atg7,PI3K,Akt and Glut4 in drug treatment-groups were improved(P<0.05).Conclusion:Improving the ability of autophagy and enhance the PI3K/Akt/GLUT4 signaling pathway may be the mechanism of Erchen decoction inhibit the level of IR of PCOS.
