摘　　要：建立高效液相色谱指纹图谱评价泻白散及其炒桑白皮、地骨皮、炙甘草3种单味药内在质量的方法。采用C_(18)色谱柱(250 nm×4.6 mm,5μm),以乙腈-0.1%甲酸水溶液为流动相,梯度洗脱,柱温为30℃,流量为1 mL/min,检测波长为254 nm,进样体积为5μL,分析时间为120 min,采用二级管阵列检测器进行测定,用高效液相色谱(HPLC)进行指纹图谱研究。结果表明,市售桑白皮饮片13个批次之间的相似度为0.024~0.975;市售地骨皮饮片13个批次之间的相似度为0.061~0.983,不同批次间差别较大;炙甘草饮片13个批次之间的相似度为0.978~0.998,相似度非常高。对14批泻白散组方的水煎液进行HPLC指纹图谱分析发现,有9个共有峰,可指认其中的地骨皮乙素、异甘草苷、甘草酸3个峰。该方法能够反映市售泻白散及其3种单味药的内在质量,为泻白散经典名方的质量标准制定及临床用药提供参考。A high performance liquid chromatography(HPLC)fingerprint method was established to evaluate the internal quality of Xiebai powder and its three single herbs:Kukoamine B,Isoliquiritin,Glycyrrhizic acid.C_(18) chromatographic column(250 nm×4.6 mm,5μm)was used with acetonitrile-0.1%formic acid aqueous solution as the mobile phase by gradient elution,the column temperature was 30℃,the flow rate was 1 mL/min,the detection wavelength was 254 nm and the injection volume was 5μL.The analysis time was 120 min.The two-stage tube array detector was used for determination,and the fingerprint was studied by high performance liquid chromatography.The results showed that the similarity between 13 batches of Kukoamine B decoction pieces was 0.024-0.975;the similarity between 13 batches of Isoliquiritin decoction pieces on the market was 0.061-0.983,and there was a great difference among different batches;the similarity between 13 batches of Glycyrrhizic acid was 0.978-0.998,which was very high.HPLC fingerprint analysis of 14 batches of decoction of Xiebai powder showed that there were 9 common peaks,including 3 peaks of Kukoamine B,Isoliquiritin,Glycyrrhizic acid.This method can reflect the internal quality of three single drugs of Xiebai powder sold in the market,and provide reference for the formulation of quality standards of classic famous prescriptions of Xiebai powder and clinical medication.
