摘 要:目的基于PI3K/AKT通路探究沉默肿瘤坏死因子受体相关分子6(TRAF6)对柯萨奇病毒性心肌炎(VMC)模型小鼠的干预效果。方法2019年9月—2020年9月于广西柳州市工人医院实验室进行实验,选取健康雄性SD小鼠44只,随机数字表法选取11只作为正常组,33只建立柯萨奇VMC模型,并随机分为模型组、上调组、沉默组各11只,上调组小鼠尾部静脉注射agoTRAF 630 mg/kg,沉默组小鼠尾部静脉注射antagoTRAF 630 mg/kg,正常组、模型组小鼠尾部静脉注射等剂量0.9%氯化钠溶液进行干预。比较各组小鼠心功能指标,CK-MB、cTnT、IL-6、TNF-α、IFN-γ、SOD、MDA水平,PI3K、AKT及TRAF6表达水平。结果与正常组比较,模型组LVESD、LVEDD升高,LVEF降低(P<0.05);与模型组比较,上调组LVESD、LVEDD升高,LVEF降低(P<0.05),而沉默组LVESD、LVEDD降低,LVEF升高(P<0.05);与上调组比较,沉默组LVESD、LVEDD降低,LVEF升高(P<0.05)。与正常组比较,模型组小鼠CK-MB、cTnT水平升高(P<0.05);与模型组比较,上调组CK-MB、cTnT水平升高,沉默组CK-MB、cTnT水平降低(P<0.05);与上调组比较,沉默组CK-MB、cTnT水平降低(P<0.05)。与正常组比较,模型组IL-6、TNF-α、IFN-γ水平升高(P<0.05);与模型组比较,上调组IL-6、TNF-α、IFN-γ水平升高,沉默组IL-6、TNF-α、IFN-γ水平降低(P<0.05);与上调组比较,沉默组IL-6、TNF-α、IFN-γ水平较低(P<0.05)。与正常组比较,模型组SOD水平降低,MDA水平升高(P<0.05);与模型组比较,上调组SOD水平降低,MDA水平升高,沉默组SOD水平升高、MDA水平降低(P<0.05);与上调组比较,沉默组SOD水平升高,MDA水平降低(P<0.05)。与正常组比较,模型组PI3K、AKT、TRAF6表达升高(P<0.05);与模型组比较,上调组PI3K、AKT、TRAF6表达升高,沉默组PI3K、AKT、TRAF6表达降低(P<0.05);与上调组比较,沉默组PI3K、AKT、TRAF6表达降低(P<0.05)。结论通过PI3K/AKT通路沉默TRAF6对柯萨奇VMC模型小鼠进行干预,可有效降低炎性反�Objective To probe the intervention effect of silencing TRAF6 based on PI3K/Akt pathway in coxsackievirus myocarditis model mice.Methods From September 2019 to September 2020,the experiment was carried out in the laboratory of Liuzhou Workers'Hospital,Guangxi.44 SD healthy male mice were selected,11 of which were considered as the normal group,and 33 mice were established Coxsackie VMC model.They were randomly divided into model group,upregulated group and silent group,and the 4 groups of mice were respectively interfered.Cardiac function indexes,levels of CK-MB,cTnT,IL-6,TNF-α,IFN-γ,SOD,MDA,expression of PI3K Akt and TRAF6 were compared in each group.Results Compared with the normal group,LVESD and LVEDD were increased and LVEF was decreased in the model group(P<0.05).Compared with model group,LVESD,LVEDD and LVEF were decreased and LVEF were increased in normal group and silent group,while LVESD,LVEDD and LVEF were increased and LVEF was decreased in up-regulated group(P<0.05).Compared with the upregulated group,LVESD,LVEDD and LVEF were decreased and increased in the normal group,model group and silent group(P<0.05).Compared with normal group,the levels of CK-MB and cTnT in model group were increased(P<0.05).Compared with model group,the levels of CK-MB and cTnT were decreased in normal group and silent group,and increased in upregulated group(P<0.05).Compared with the upregulated group,the levels of CK-MB and cTnT in normal group,model group and silent group were lower(P<0.05).Compared with normal group,the levels of IL-6,TNF-αand IFN-γin model group were increased(P<0.05).Compared with model group,the levels of IL-6,TNF-αand IFN-γin normal group and silent group were decreased,while the levels of IL-6,TNF-αand IFN-γin upregulated group were increased(P<0.05).Compared with the upregulated group,the levels of CK-MB and cTnT were lower in the normal group,model group and silent group(P<0.05).Compared with normal group,SOD level in model group was decreased and MDA level was increased(P<0.05).Compared wi
