摘　　要：【目的】研究胚胎着床期瘦素(leptin)及其受体(OB-R)在真孕和假孕母犬下丘脑-垂体-性腺轴各组织中的表达差异.【方法】应用反转录聚合酶链反应(RT-PCR)和免疫组织化学的方法检测胚胎着床期真孕和假孕母犬下丘脑、垂体、卵巢、输卵管、子宫组织中瘦素及其受体的表达.【结果】瘦素在真孕和假孕母犬的下丘脑、垂体、卵巢、子宫中均有表达,在输卵管中表达阴性,瘦素受体在真孕和假孕母犬各组织中均有表达;瘦素及其受体在真孕母犬下丘脑、卵巢和子宫组织中的表达量显著高于假孕母犬(P<0.05),瘦素受体在真孕母犬垂体和输卵管中的表达量显著高于假孕母犬(P<0.05).真假孕母犬垂体中瘦素水平差异不显著(P>0.05).【结论】瘦素及其受体对于犬的胚胎着床具有一定调节作用.【Objective】To study the expression differentiation of leptin and its receptor(OB-R)in various tissues of hypothalamus-pituitary-gonad axis during embryo implantation.【Method】Reverse transcription polymerase chain reaction(RT-PCR)and immunohistochemistry assays were used to determine the expression differentiation of leptin and the receptor in various tissues of hypothalamus-pituitary-gonad axis.【Result】Leptin was expressed in hypothalamus,pituitary,ovary and uterus of pregnant and pseudo-pregnant bitches except fallopian tube.Meanwhile,Leptin receptor was expressed in all the above five tissues of pregnant and pseudo-pregnant bitches at implantation stage.The expression levels of leptin and its receptor in hypothalamus,ovary and uterus of pregnant bitches were significantly higher than that of pseudo-pregnant bitches(P<0.05).Moreover,the expression level of leptin receptor in pituitary and fallopian tube of pregnant bitches was dramatically higher than that of pseudo-pregnant bitches(P<0.05).However,there was no significant expression differentiation of leptin in pituitary of pregnant and pseudo-pregnant bitches(P>0.05).【Conclusion】Leptin and its receptor exert a certain role in regulating the embryo implantation of bitches.
