摘  要:目的研究微RNA(miR)-146a-5p靶向白细胞介素-1受体相关激酶1(IRAK-1)调控深静脉血栓(DVT)患者内皮祖细胞(EPCs)的增殖及炎症。方法选取2018年7月至2019年6月该院诊断为DVT的10例患者及10例体检的健康志愿者为研究对象,分离外周血EPCs,检测miR-146a-5p、IRAK-1的表达水平;DVT患者的EPCs分为miR-阴性对照(NC)组、miR-146a-5p组、miR-NC+NC质粒组、miR-146a-5p+NC质粒组、miR-146a-5p+IRAK-1质粒组,检测细胞增殖活力及吸光度(A490)值、miR-146a-5p、IRAK-1、核因子-κB(NF-κB)、肿瘤坏死因子-α(TNF-α)、白细胞介素-1β(IL-1β)水平。结果DVT患者EPCs中miR-146-5p相对表达水平低于健康志愿者,IRAK-1相对表达水平高于健康志愿者(P<0.05);miR-146a-5p组A490值、miR-146a-5p相对表达水平高于miR-NC组,IRAK-1、NF-κB、TNF-α、IL-1β表达水平、野生型IRAK-1基因的荧光活力低于miR-NC组(P<0.05);miR-146a-5p+IRAK-1质粒组A490值低于miR-146a-5p+NC质粒组,IRAK-1、NF-κB、TNF-α、IL-1β水平高于miR-146a-5p+NC质粒组(P<0.05)。结论miR-146a-5p靶向IRAK-1促进DVT患者的EPCs增殖并抑制炎性反应。Objective To study the effect of microRNA(miR)-146a-5p on the proliferation and inflammation of endothelial progenitor cells(EPCs)of deep vein thrombosis(DVT)patients by targeting interleukin-1 receptor associated kinase 1(IRAK-1).Methods A total of 10 patients with DVT and 10 healthy volunteers in hospital from July 2018 to June 2019 were selected.EPCs from peripheral blood were isolated and the expression levels of miR-146a-5p and IRAK-1 were detected.EPCs of DVT patients were divided into the miR-negative control(NC)group,the miR-146a-5p group,the miR-NC+NC plasmid group,the miR-146a-5p+NC plasmid group,the miR-146a-5p+IRAK-1 plasmid group.The proliferation viability A490 vale,the expression levels of miR-146a-5p,IRAK-1,NF-κB,the contents of tumor necrosis factor-α(TNF-α)and interleukin-1β(IL-1β)were measured.Results The expression level of miR-146-5p in EPCs of DVT patients was lower than that of healthy volunteers,and the expression level of IRAK-1 was higher than that of healthy volunteers(P<0.05).A490 value and the expression level of the miR-146a-5p group and the miR-146a-5p group were higher than those of the miR-NC group,the expression levels of IRAK-1,NF-κB,the contents of TNF-α,IL-1βand fluorescence activity of wild-type IRAK-1 gene were lower than those of the miR-NC group(P<0.05).A490 value of miR-146a-5p+IRAK-1 plasmid group were lower than those of the miR-146a-5p+NC plasmid group,the expression levels of IRAK-1,NF-κB,the contents of TNF-α,IL-1βwere higher than those of the miR-146a-5p+NC plasmid group(P<0.05).Conclusion MiR-146a-5p promotes proliferation and inhibits inflammation of EPCs of DVT patients by targeting IRAK-1.
