摘 要:目的观察电针不同腧穴对功能性便秘小鼠肠道动力及近端结肠组织脑源性神经营养因子(BDNF)和乙酰胆碱转移酶(ChAT)表达的影响,探讨其作用机制。方法14 d冷饮刺激制备功能性便秘小鼠模型。实验小鼠随机分为正常组、模型组、上巨虚组和天枢组。上巨虚组和天枢组使用电针刺激,参数为2/15 Hz,1 mA,15 min/d,共3 d。治疗结束后,观察首粒红便时间,Western blot检测小鼠近端结肠组织BDNF和ChAT蛋白表达,电生理实验观察近端结肠肠内压改变。结果与正常组比较,模型组小鼠首粒红便时间明显延长(P<0.01),近端结肠组织BDNF和ChAT蛋白表达明显降低(P<0.05,P<0.01)。与模型组比较,上巨虚组小鼠首粒红便时间明显缩短(P<0.01),与正常小鼠无明显差异,近端结肠肠内压明显升高(P<0.05),BDNF和ChAT蛋白表达明显上调(P<0.05,P<0.01),且恢复至正常水平;天枢组小鼠首粒红便时间明显缩短(P<0.05),但与上巨虚组和正常组比较差异有统计学意义(P<0.05),近端结肠肠内压和BDNF、ChAT蛋白表达无明显变化。结论电针“上巨虚”“天枢”均能缓解小鼠便秘症状,但在近端结肠部位其作用不同。Objective To observe the effects of electroacupuncture at different acupoints points on the intestinal motility and the expression of BDNF and ChAT in proximal colon tissue of mice with functional constipation,and to explore the action mechanism.Methods The model of functional constipation was established by cold drink stimulation for 14 days.The mice were randomly divided into normal group,model group,Shangjuxu group and Tianshu group.Electroacupuncture was used in Shangjuxu group and Tianshu group with parameters of 2/15 Hz,1 mA and time of 15 min/d for 3 days.After treatment,the first defecation time was observed and the expression of BDNF and ChAT protein in proximal colon tissue was analyzed by Western blot.Electrophysiological experiment was used to observe the changes of proximal colonic pressure.Results Compared with the normal group,the time of first defecation in mice of model group was extended(P<0.01),the BDNF and ChAT protein expression in proximal colon tissue were significantly decreased(P<0.05,P<0.01).Compared with the model group,the time of first defecation in mice of Shangjuxu group was shorter(P<0.01),and there was no significant difference compared with the normal group;the intestinal pressure in the proximal colon increased(P<0.05);the expression of BDNF and ChAT protein were significantly up-regulated(P<0.05,P<0.01),and return to normal levels.The time of first defecation in mice of Tianshu group was significantly shortened(P<0.05),but it was significantly different from that in Shangjuxu group and normal group(P<0.05);the intestinal pressure,BDNF and ChAT expression in the proximal colon did not change significantly.Conclusion Electroacupuncture at“Shangjuxu”“Tianshu”can alleviate constipation symptoms,but their effects are different in the proximal colon.
