摘 要:目的研究蒙药古日古木-13对光损伤小鼠视网膜细胞组织结构及功能的影响,评价其抑制细胞凋亡的能力及对小鼠视网膜光损伤的保护作用。方法40只7~8周龄SPF级雄性C-57小鼠,随机分为5组:正常组、对照组和治疗低、中、高剂量组,每组8只。所有小鼠进行1 d暗适应后,在试验用光损伤箱中进行冷照射,照射结束后送返至暗环境12 h,光照后灌胃给药7 d,通过视网膜电图(ERG)评估各组小鼠视网膜功能变化、光学显微镜观察视网膜形态学变化、Western印迹检测凋亡蛋白含半胱氨酸的天冬氨酸蛋白水解酶(caspase)-3、caspase-8、caspase-9、B细胞淋巴瘤(Bcl)-2、Bcl-2相关X蛋白(Bax)表达。结果治疗组ERG波形振幅数值显著大于对照组数值,差异有统计学意义(P<0.001);光镜观察对照组视网膜外核层厚度与正常组相比明显减小,而治疗组较对照组视网膜外核层厚度稍有增大,差异有统计学意义(P<0.05);Western印迹检测Caspase-3、8、9、Bax蛋白表达量低于对照组,Bcl-2蛋白表达量高于对照组,差异有统计学意义(P<0.05)。结论蒙药古日古木-13对光损伤小鼠视网膜组织结构及功能有保护作用,对光损伤小鼠视网膜细胞凋亡有抑制作用。Objective To study the effect of GRGM-13 on the repair of retinal cell structure and function in light-damaged mice.To assess the inhibition of GRGM-13 on cell apoptosis and the protective effect on retinal light injury in mice.Methods 40 SPF grade male C-57 mice aged 7~8 weeks were randomly divided into five groups,including normal group,control group,high dose GRGM-13 group[1.6 g/(kg•d)],middle dose GRGM-13 group[0.8 g/(kg•d)],low dose GRGM-13 group[0.4 g/(kg•d)],eight mice in each group.After dark adaptation for 1 day,all mice were subjected to cold irradiation,then they were sent back to dark environment for 12 h and given by gavage for 7 days.After the changes of retinal function were assessed by ERG,the morphological alterations of retina were observed by light microscope and detected the expressions of apoptotic protein caspase 3,8,9 and Bax/Bcl-2 by Western blot.Results ERG showed that waveform amplitude value in the Mongolian medicine group was significantly greater than that of the control group,and the difference was statistically significant(P<0.001).Microscopic examination results showed that the thickness of the outer nuclear layer in the control group was significantly decreased compared with the normal group,while the thickness of the outer nuclear layer in the Mongolian medicine group was slightly increased compared with the control group,and the difference was statistically significant(P<0.05).Compared with control group,Western blot showed the expressions of caspase-3,8,9 and Bax protein in the Mongolian medicine group were significantly lower but Bcl-2 protein was significantly higher(P<0.05).Conclusions Mongolian medicine GRGM-13 has a protective effect on the retinal structure and function of light-damaged mice.Mongolian medicine GRGM-13 has an inhibitory effect on the apoptosis of retinal cells in light-damaged mice.
