斑蝥酸钠注射液靶向PI3K/AKT/mTOR信号通路对人肺腺癌细胞A549迁移能力的影响

摘　　要：目的:探讨斑蝥酸钠注射液对人肺腺癌细胞A549侵袭与转移的影响及其作用机制。方法:分别用0.312 5 mg·L^(-1)、0.625 mg·L^(-1)、1.25 mg·L^(-1)、2.5 mg·L^(-1)、5 mg·L^(-1)的斑蝥酸钠注射液处理A549细胞,MTT法检测斑蝥酸钠注射液对A549细胞增殖的影响,并计算半数抑制浓度(half maximal inhibitory concentration, IC50);根据MTT结果,在后续实验中,将细胞分为对照组(control)、斑蝥酸钠注射液低剂量组、斑蝥酸钠注射液高剂量组及顺铂组(DDP),通过细胞划痕实验检测斑蝥酸钠注射液对A549细胞侵袭与转移能力的影响;Western Blot检测斑蝥酸钠注射液对A549细胞磷脂酰肌醇3激酶/蛋白激酶B/哺乳动物雷帕霉素靶蛋白(phosphatidylinositol 3 kinases/protein kinase B/mammalian target of rapamycin, PI3K/AKT/mTOR)通路相关蛋白表达的影响。结果:斑蝥酸钠注射液显著提高A549细胞的增殖抑制率(P<0.05),且呈浓度及时间依赖性;与control组比较,不同剂量的斑蝥酸钠注射液能显著降低A549细胞的迁移率及PI3K、AKT、mTOR、P70 S6和VEGF-C的蛋白表达水平,且呈浓度依赖性。结论:斑蝥酸钠注射液能显著抑制肺癌A549细胞的增殖和迁移,作用机制可能与其抑制PI3K/AKT/mTOR通路及血管生成有关。Objective: To investigate the effect of sodium cantharidate injection on the invasion and metastasis of human adenocarcinoma cancer cell A549 and its mechanism.Methods: Lung cancer A549 cells were treated with sodium cantharidate injection of 0.312 5 mg·L^(-1),0.625 mg·L^(-1),1.25 mg·L^(-1),2.5 mg·L^(-1) and 5 mg·L^(-1) respectively.The effect of sodium cantharidate injection on the proliferation of lung cancer A549 cells was detected by MTT method, and the half maximal inhibitory concentration(IC50) was calculated;according to the MTT results, in the follow-up experiment, the cells were divided into control group, low-dose group, high-dose group and cisplatin(DDP) group.The effect of sodium cantharidate injection on the invasion and metastasis of lung cancer A549 cells was detected by cell scratch test;the effect of sodium cantharidate injection on the expression of phosphatidylinositol 3 kinases/protein kinase B/mammalian target of rapamycin PI3 K/AKT/mTOR pathway related proteins in lung cancer A549 cells was detected by Western Blot.Results: Sodium cantharidate injection significantly increased the cell proliferation inhibition rate of A549 cells(P<0.05) in a concentration and time-dependent manner;compared with the control group, different doses of sodium cantharidate injection could significantly reduce the migration rate of A549 cells and the protein expression of PI3 K,AKT,mTOR,P70 S6 and VEGF-C in a concentration dependent manner.Conclusion: Sodium cantharidate injection can significantly inhibit the proliferation and migration of lung adenocarcinoma A549 cells, and its mechanism may be related to its inhibition of the activation of PI3 K/AKT/mTOR pathway and angiogenesis.
