益气温阳方联合顺铂对Lewis荷瘤小鼠抗肿瘤的作用及对Th1/Th2平衡的影响

摘　　要：目的:观察益气温阳方联合顺铂对Lewis荷瘤小鼠的抗抑瘤作用及对辅助性T淋巴细胞1(T-helper 1,Th1)、辅助性T淋巴细胞2(T-helper 2,Th2)相关细胞因子的影响。方法:将32只雄性C57BL/6小鼠随机分为模型组、益气温阳方组(20 g·kg^(-1))、顺铂组(4 mg·kg^(-1))及联合用药组(益气温阳方+顺铂),每组8只。采用皮下注射Lewis细胞建立荷瘤小鼠模型,连续给予相应的药物12 d。检测小鼠的抑瘤率;ELISA法检测血清白细胞介素-2(interleukin-2,IL-2)、白细胞介素-4(interleukin-4,IL-4)、白细胞介素-10(interleukin-10,IL-10)、干扰素-γ(interferon-γ,IFN-γ)水平;HE染色观察小鼠肿瘤组织病理变化。结果:与模型组比较,益气温阳方组、顺铂组及联合用药组均有显著的抑瘤作用,可显著升高血清IL-2、IFN-γ水平(P<0.01),显著降低IL-4、IL-10水平(P<0.01),可见明显坏死区域,免疫细胞浸润增加。结论:益气温阳方可通过调节Th1/Th2的免疫失衡状态发挥抗肿瘤的作用。Objective: To observe the anti-tumor effect of Yiqi Wenyang Prescription combined with cisplatin on Lewis tumor bearing mice and the effects on T-helper 1(Th1) and T-helper 2(Th2) related cytokines.Methods: 32 male C57 BL/6 mice were randomly divided into four groups: model group, Yiqi Wenyang Prescription group(20 g·kg^(-1)),cisplatin group(4 mg·kg^(-1)) and combined medication group(Yiqi Wenyang Prescription + cisplatin),with 8 mice in each group.The tumor bearing mouse model was established by subcutaneous injection of Lewis cells, and the corresponding drugs were given continuously for 12 days.The tumor inhibition rate of mice was detected;ELISA method was used to detect serum interleukin-2(IL-2),interleukin-4(IL-4),interleukin-10(IL-10),interferon-γ(IFN-γ) level;HE staining was used to observe the pathological changes of tumor tissue in mice.Results: Compared with the model group, Yiqi Wenyang Prescription group, cisplatin group and combined drug group had significant antitumor effects, and could significantly increase serum IL-2 and IFN-γ.The levels of IL-4 and IL-10 were significantly reduced(P<0.01).Obvious necrotic areas and increased immune cell infiltration were observed.Conclusion: Yiqi Wenyang Prescription can play an anti-tumor role by regulating the immune imbalance of Th1/Th2.
